丹参酮Ⅱ_A对高糖透析诱导小鼠腹膜纤维化的影响

【摘要】 目的研究中药丹参化学成分丹参酮ⅡA(TanshinoneⅡA,TⅡA)对小鼠腹膜纤维化模型的影响。方法 50只小鼠随机分为空白对照组、模型组、TⅡA低剂量组、TⅡA中剂量组与TⅡA高剂量组,每组10只动物。模型组与TⅡA组连续腹腔注射腹透液30d,TⅡA组同时腹腔注射不同剂量丹参酮ⅡA磺酸钠。取腹壁组织、血清与腹腔透析液,分别检测羟脯氨酸含量、腹透1hr透析液/透析液起始葡萄糖浓度比值(D1/D0GLU)与透析液/血清尿素浓度比值(D/P BUN),比较各组之间的差异。结果与空白对照组相比,模型组腹壁羟脯氨酸含量增加,D/P BUN值明显升高,D1/D0GLU值降低,统计学差异显著(P<0.01),说明腹膜纤维化模型制备成功。TⅡA中、高剂量组羟脯氨酸含量均低于模型组,统计学差异显著(P<0.01),与空白对照组相比无统计学差异(P>0.05)。TⅡA中、高剂量组D1/D0GLU值均高于模型组,统计学差异显著(P<0.01)。TⅡA低、中、高剂量组D/P BUN值虽比模型组有所降低,但无统计学差异(P>0.05)。由此推断丹参酮ⅡA具有改善腹膜功能以及减少胶原沉积的作用。结论丹参酮ⅡA为丹参预防腹膜纤维化的有效成分。

【Abstract】 OBJECTIVE To study the effect of tanshinone ⅡA(T ⅡA) on peritoneal fibrosis(PF) in mice. METHODS 50 mice were randomly divided into 5 groups: blank control group,model group,TⅡ Alow dosage group,TⅡ Amedium dosage group,TⅡ Ahigh dosage group(n = 10 in each group). Intraperitoneal injection of peritoneal dialysate were performed in model group and TⅡ Agroups for 30 d. TⅡ Agroups were also administered with various dosages of tanshione ⅡAsulfonic Sodium. Afterwards,peritoneal tissue,serum and dialysis fluid were collected to detect the contents of hydroxyproline,the glucose concentration ratio of dialysis fluid that dialysed for 1hr to the inital dialysis fluid(D1/ D0GLU),and the concentration ratio of dialysate to serum urea(D/P BUN). At last,compare the difference between groups. RESULTS Compared with blank group,the model groups had the higher hydroxyproline content,the significantly higher D/P BUN and the lower D1/ D0GLU(P < 0. 01) Comparison of groups indicated that the preparation of PF model was successful. TⅡ Amedium dosage and TⅡ Ahigh dosage group had lower hydroxyproline content than the model group(P < 0. 01),but there were no significant difference with the blank group(P > 0. 05). TⅡ Amedium dosage and TⅡ Ahigh dosage group were higher D1/ D0 GLU than the model group(P < 0. 01). Although T ⅡAgroups were lower D/P BUN than the model group,there were no significant difference in TⅡ Agroups and model group(P > 0. 05). Thus tanshinone ⅡAcan improve the function of peritoneum and reduce the effect of collagen deposition. CONCLUSION TⅡ Ais the active constituent of Salvia Miltiorrhiza for PF prevention.
