气相色谱法测定当归挥发油中藁本内酯的含量

【摘要】 目的建立当归挥发油中藁本内酯的含量测定方法。方法采用水蒸气蒸馏法进行挥发油的提取,以液体石蜡为吸收剂、无水乙醇为萃取剂,采用GC法对挥发油藁本内酯进行含量测定。气相色谱条件:AT-SE-54毛细管柱(15m×0.25mm×0.33μm);氢焰离子化检测器(FID);进样口温度230℃;检测器温度250℃;柱温以160℃为起始温度,保持2min,以10℃/min升温至200℃,保留2min。载气为氮气。进样量为1μL。结果在101.5μg·mL-1~609.0μg·mL-1范围内呈良好线性关系,峰面积与浓度的回归方程为:Y=522.23X+5882.6(R2=0.999),精密度试验RSD:1.76%,重复性试验RSD:0.70%,平均加样回收率96.49%。结论方法简便快速,试剂安全无毒,可用于当归挥发油中藁本内酯的含量测定。

【Abstract】 OBJECTIVE To establish a GC method for determination ligustilide in volatile fractions of Angelica sinensis( Oliv.) Diels. METHODS The sential oil was extracted by steam distillation. Using liquid paraffin as absorbent,anhydrous ethanol as extractant. GC method was applied for the determination of ligustilide from the essential oil. Chromatographic conditions: Capillary column AT-SE-54( 15 m × 0. 25 mm × 0. 33μm) was used; The detectorwas FID; Inlet temperature was 230℃; The detector temperature was 250℃; Column temperature: 160℃( 2min) 10℃ /min 200℃( 2min); Carrying gas was nitrogen; Injection volume was 1μL; RESULTS There is a good linear relationshipin in 101. 5μg·mL- 1~ 609μg·mL- 1range,the peak area and the concentration of the regression equation:Y = 522. 23 X + 5882. 6( R2= 0. 9990),precision test RSD was 1. 76%,reproducible test RSD was 0. 70%,the average recovery rate was 96. 49%. CONCLUSION This method is simple,quick and specific,reagent is safe and nontoxic. It can be applied for determination ligustilide in volatile fractions of Angelica sinensis( Oliv.) Diels.
