油酸诱导急性呼吸窘迫综合征大鼠心脏局部RAS的影响及心功能变化相关研究

【摘要】 目的观察油酸诱导急性呼吸窘迫综合征(ARDS)大鼠心脏局部RAS的影响及心功能的变化。方法雄性SD大鼠36只,随机分成假手术组和造模组,每组18只,各组又按造模成功后1h、2h和4h分为a、b、c 3个亚组。(1)假手术组(S组):仅进行气管切开插管和右颈总动脉穿刺置管,尾静脉注射生理盐水0.15mL/kg;(2)造模组(C组):尾静脉注射油酸0.15mL/kg,建立油酸诱导的大鼠ARDS模型。每组有效检测与活杀取材的大鼠均为18只。结果与S组比较,C组LVEDP、AngⅡ含量及Renin活性均明显升高(P<0.05),LVSP、±LVdp/dtmax明显下降。结论 ARDS可致大鼠心肌损害、心功能降低,同时激活心脏局部RAS。

【Abstract】 Objective To observe the relationship of the cardiac function and the level of local RAS in acute respiratory distress syndrome(ARDS)rats induced by oleic acid.Methods Sprague-Dawley rats were randomly divided into two groups.Group S:Control Group(only tracheal intubation,the right carotid artery catheterization,and tail vein injection of normal saline 0.15mL/kg.n=18);Group C(ARDS group:the ARDS model of rats induced by tail vein injection of oleic acid 0.15mL/kg,n=18).Two groups were randomly divided into 3subgroups respectively by 1h,2hand 4hafter the modeling,and there were6 rats in each group with effective detection and killed alive for materials.Recording the left ventricular systolic pressure(LVSP),maximum rising and falling rate of left intraventricular pressure(±LVdp/dtmax),and left ventricular end-diastolic pressure(LVEDP)at each time point.AngiotensinⅡ(AngⅡ)concentration in myocardial tissue and renin activity level(Renin)were be detected respectively.Results In ARDS rats,LVSP,LVEDP,± LVdp/dtmax,AngⅡ content and Renin activity were higher than control group with significant difference(P<0.05).ConclusionARDS could induce myocardial damage,decrease cardiac function and activation of cardiac local RAS in rats..
