健脾消积汤抑制二乙基亚硝胺致大鼠肝癌前病变的作用

【摘要】 目的观察健脾消积汤抑制二乙基亚硝胺(DEN)致大鼠肝癌前病变的作用及其机制。方法实验用雄性SD大鼠80只,按抽签法随机分为4组(大、小剂量中药组及模型组、空白组),每组20只。除空白组正常饲养外,其余3组均用DEN作为肝癌的启动剂(200μg/kg),然后喂食含0.015%的2-乙酰氨基芴(2-AAF)饲料2周,行大部分肝切除术。大、小剂量中药组大鼠从实验开始即加用健脾消积方药汤剂灌胃,模型组则单纯喂食基础饲料。于停饲2-AAF饲料3 d后抽取大鼠血液并将其处死,检测大鼠血清白介素2(IL-2)含量,肝脏按规定部位取材,ABC法行转化生长因子β1(TGF-β1)免疫组织化学染色,另外肝脏采用γ-谷氨酰转肽酶(γ-GT)染色,算出平均每平方厘米肝面积中γ-GT阳性灶的数量、面积。结果大、小剂量中药组γ-GT阳性灶的数量、面积均明显少于模型组(P<0.05),大、小剂量中药组间无明显差异(P>0.05)。免疫组化检测显示小剂量中药组TGF-β1阳性率较模型组低(P<0.05),大剂量中药组TGF-β1阳性率低于模型组,但差异无统计学意义(P>0.05)。大剂量中药组、小剂量中药组、模型组大鼠血清IL-2含量均较空白组降低(P<0.05),大剂量中药组、小剂量中药组降低程度较模型组低(P<0.05),大剂量中药组、小剂量中药组IL-2含量无显著差异(P>0.05)。结论健脾消积方药有一定的抑制大鼠肝癌前病变作用,其机理可能是能过抑制TGF-β1生成,降低对免疫因子IL-2的抑制作用,增强大鼠免疫功能而起作用。

【Abstract】 Objective To observe the effects of Jianpi Xiaoji Tang on precancerous lesions in rats with diethylnitrosamine( DEN)-induced liver cancer and its mechanism. Methods Eighty laboratory SD male rats were randomly divided into four groups according to drawing lots,low-dosage traditional Chinese medicine( TCM) group,high-dosage TCM group,model group and blank group,20 rats in each group. Except the rats in the blank group were given normal breeding,the rats in other 3 groups were administered with the liver cancer initiator of DEN( 200 μg / kg),and then with food containing 0. 015% of 2-acetylaminofluorene( 2-AAF) for 2 weeks,following partial hepatectomy. On the beginning of this experiment,the rats in the high-dosage and low-dosage TCM groups were given Jianpi Xiaoji Tang by gavage,and the rats in the model group were only fed with normal food. On the third day after stopping the feeding of 2-AAF-contained food,the rats were put to death following their blood were drew for the detection of the serum levels of interleukin-2( IL-2).The targeting samples of liver were collected,ABC method was applied to immunohistochemical staining for determination of transforming growth factor β1( TGF-β1),the samples of liver were stained by γ-glutamyltranspeptidase( γ-GT),and the numbers and areas of γ-GT positive foci per cm2of the liver area were calculated. Results The numbers and areas of γ-GT positive foci in the high-dosage and low-dosage TCM groups were significantly less than those of model group( P <0. 05),no significant difference exsisted between low-dosage TCM group and high-dosage TCM group( P > 0. 05). Immunohis-tochemical detection showed that the positive rate of TGF-β1 in the low-dosage TCM group was significantly lower than that of model group( P < 0. 05),which was lower in the high-dosage TCM group in contrast with that of model group but showed no significant difference( P > 0. 05). The serum levels of IL-2 decreased significantly in the high-dosage and low-dosage TCM groups,model group in contrast with that in the blank group( P < 0. 05),and the decrease in the high-dosage and low-dosage TCM groups was more significant than that in the model group( P < 0. 05),there was no significant difference in the level of IL-2 between high-dosage and low-dosage TCM groups( P > 0. 05). Conclusion Jianpi Xiaoji Tang has some effects on inhibiting precancerous lesion in rats with liver cancer. Its mechanism might relate with its effects of inhibiting TGF-β1’s generation,reducing the inhibition of IL-2 and enhancing the immune function of rats.
