纤溶酶原激活物与基质金属蛋白酶在矽肺纤维化中的作用
The mechanism of uPA and MMP-9 in pneumonoconiosis
【摘要】 目的研究纤溶酶原激活物(uPA)与基质金属蛋白酶(MMP-9)在大鼠矽肺纤维化发生发展中的意义。方法建立大鼠矽肺模型组及对照组,分别取不同时间点处死大鼠,取其肺组织,免疫组化法检测肺组织中uPA、MMP-9蛋白的表达并分析其相关性。结果与对照组比较,uPA的阳性表达随时间延长呈增强趋势,第1周时达高峰,此后逐渐减弱,但仍较对照组明显。MMP-9阳性表达强度随染尘时间延长而逐渐增强,至第7天时达高峰,此后呈减弱趋势。uPA、MMP-9蛋白表达与对照组比较,差异有统计学意义(P<0.05)。其显色指数与对照组比较,差异有统计学意义(P<0.05)。uPA、MMP-9蛋白表达呈正相关。结论肺组织内uPA、MMP-9蛋白表达在肺纤维化过程中发挥重要的作用,可能因为uPA诱导的MMPs活化与其纤溶活性共同改变了细胞外基质胶原的沉积,从而影响矽肺纤维化的发生发展。
【Abstract】 Objective To observe the expression of uPA and MMP-9in silicosis in rats in order to investigate the mechanism in Pneumonoconiosis.Methods Wistar rats were randomly divided into normal control group and silicosis-induced group.After establishing the silicosis model in rats,the rats from each group were sacrificed at different time.The expressions of uPA and MMP-9in the lung tissues of the rats were detected.Results Compared with that in the control group,the expression of uPA in the silicosis-induced group increased significantly;while MMP-9increased gradually,and reached highest level at 7d,then reduced but significantly higher than normal.Compared with control group,the color rendering Index of uPA and MMP-9were higher significantly.Conclusions There were positive relationship among uPA and MMP-9;the enhance of MMPs may be correlated with uPA;uPA and MMPs promote the silicosis induced lung fibrosis together.
