碱茅(Puccinellia tenuifolra)全长cDNA酵母文库中与铝胁迫相关基因的初步筛选
The Full-length cDNA Library with Yeast Screening of Resistant Aluminum-related Genes from Puccinellia tenuifolra
【摘要】 利用碱茅全长cDNAs酵母表达文库,经不同浓度AlCl3筛选得到19个不同克隆可增强酵母的耐铝性,经过同源性比对后,将这19个对Al3+胁迫敏感的抗性克隆大体分为以下几类:结构功能蛋白、酶类、有跨膜结构的蛋白、与光合作用相关蛋白以及功能未知蛋白。为了了解该19个单克隆对于胁迫的耐受特异性,本研究中将这19个克隆与转空载体的酵母分别在以下各种胁迫条件下进一步进行解析:AlCl3、NaCl、H2O2。结果显示大部分筛选所得克隆对AlCl3在浓度为5.5 mmol/L时表现出耐受性。在NaCl、H2O2条件下不同克隆之间表现出不同的耐性。
【Abstract】 The Puccinellia tenuifolra cDNA libraries were expressed in yeast(Saccharomyces cerevisiae) and screened on agar plates containing different toxic concentrations of aluminum. Ninteen cDNAs were isolated that enhanced the aluminum tolerance of yeast. Through homologous BLAST, they were devided into five types:Structural and functional proteins, enzymes, proteins with transmembrane helix, proteins related to photosynthesis,and proteins with unknown functions. This 19 clones were spoted respectively on the YPG medium containing AlCl3, NaCl, H2O2. The result showed that most of these clones had resistance to AlCl3 at 5.5 mmol/L that it exhibit different resistance under NaCl, H2O2.
