重庆及周边部分地区柑橘炭疽病菌鉴定
Identification of citrus anthracnose pathogens in Chongqing and some adjacent areas
【摘要】 【目的】探索重庆及周边地区柑橘炭疽病菌种类、侵染特性,为柑橘炭疽病防控提供基础。【方法】采集重庆及周边部分地区柑橘炭疽病样本,进行分离培养、形态学观察、致病性测定和分子鉴定(rDNA转录间隔区(ITS)PCR特异性扩增、谷氨合成酶基因(GS)RFLP,及序列分析)。【结果】共获得18个炭疽菌菌株,所有菌株接种夏橙果实均可引起病斑。其中17个属于灰色快生型(FGG),鉴定为胶孢炭疽菌,仅1个属于橘红慢生型(SOG),鉴定为尖孢炭疽菌;谷氨合成酶DNA片段(GS)构建系统进化树表明胶孢炭疽菌的种群内部存在较大分化。【结论】重庆及周边地区柑橘炭疽病菌以胶孢炭疽菌为主,也存在尖孢炭疽菌(来自四川广安龙安柚),可能会引起柑橘贮藏期腐烂。
【Abstract】 【Objective】To explore the species of citrus anthracnose pathogens in Chongqing and adjacent areas,and to provide basis for prevention and control.【Method】samples of citrus anthracnose were collected from Chongqing and some adjacent Areas,and isolates were identified based on morphological characteristics of conidia and hyphae,PCR amplification of internal transcribed spacer(ITS) by using specific primer,1 kB glutamine synthetase gene RFLP and sequence analysis.【Result】18 strains wereisolated from the samples,and all of them could cause disease by inoculating on the surface of citrus fruit peel.Most of them belonged to type of Fast Growing Gray type,identified as C.gloeos porioides.Only one belonged to Slow Growing Orange type,identified as C.acutatum.Phylogenetic tree based on glutaminesyn thetase gene(GS)sequence of isolates showed obvious difference in C.gloeosporioides.【Conclusion】C.gloeosporioides is the dominant species in all areas and C.acutatum found on longan pomelo in Guangan city,Sichuan province may cause rot of citrus fruit during storage.
