刺葡萄花粉形态观察
Observation of the pollen morphology of Vitis davidii
【摘要】 【目的】探讨野生刺葡萄(Vitis davidii Foёx)的遗传多样性、各类型之间的亲缘关系及演变关系。【方法】利用扫描电镜对25份野生刺葡萄的花粉形态特征进行系统观察和分析。【结果】除供试材料澧县雌能花花粉形状不规则外,其他材料花粉粒为超长圆球形和长圆球形;两性花与雄能花具3条萌发沟;花粉粒极轴长变化为23.05~31.95μm,变异系数7.71%,赤道轴长变化为12.85~20.60μm,变异系数10.36%;花粉外壁极面纹饰呈穴状、网状、疣状及2者间的过渡状态。【结论】基于花粉粒由大到小的演化顺序,将刺葡萄各类型分为3类;刺葡萄花粉粒极面纹饰的演化趋势为:穴状-网状-疣状。刺葡萄花粉极面外壁纹饰的差异与花粉粒大小可作为研究其遗传多样性、亲缘性关系与演变进程的依据。
【Abstract】 【Objective】 The objective of the study was to explore genetic diversity, relationship and evolution between Vitis davidii Foёx clones. 【Method】 The pollen morphology was observed and analyzed by scanning electron microscope. 【Result】 The results showed that the pollen shape of V. davidii clone Lixian(♀) was irregular, while the pollen polar surface of other clones were nearly prolate or perprolate with 3colporate. Pollen polar axis length ranged from 23.05 to 31.95 μm, the coefficient of variation was 7.71%,the equatorial axis length ranged from 12.85 to 20.60 μm, the coefficient of variation was 10.15%. The extine ornamentations of pollens could be divided into three groups: cavity ornamentation, reticulate ornamentation and verrucous ornamentation. 【Conclusion】 V. davidii species can be divided into three groups.The polar exine ornamentation evolve from cavity to reticulate to verrucous. The difference of exine ornamentation and pollen morphology can provide a basis for genetic diversity, relationship and evolution between clones.
