改性氯甲基壳聚糖Ce(Ⅲ)树脂对牛血清蛋白水解作用的研究
Effect of modified merrifield-chitosan-resin complexed with Ce(Ⅲ) on bovine serum albumin hydrolysis
【摘要】 以负载1,4,7,10-四氮杂环十二烷(cyclen)的氯甲基壳聚糖树脂(merrifield chitosan resin-supported cyclen,MCRC)为载体,配合Ce(Ⅲ),得到以Ce(Ⅲ)为催化中心的改性氯甲基壳聚糖Ce(Ⅲ)树脂(MCRC-Ce(Ⅲ))。通过茚三酮比色法、SDS-PAGE电泳法及氨基酸自动分析仪等技术,检测MCRC-Ce(Ⅲ)对牛血清蛋白(BSA)的水解能力。结果表明,MCRCCe(Ⅲ)在生理pH值,60℃条件下,可将BSA水解为短肽和氨基酸,24 h后水解率可达88%,水解速率表观常数Kobs为7.05×10-2mol·L-1·h-1,重复使用6次后,仍能保持80%以上的水解能力,具有广阔的应用前景。
【Abstract】 Merrifield chitosan resin-supported cyclen( MCRC) was prepared by attaching 1,4,7,10-tetraazacyclododecane( cyclen) to Merrifield chitosan resin( MCR). Then coordination with Ce( Ⅲ) as catalytic center to be MCRC-Ce( Ⅲ). Ninhydrin reaction,SDS-PAGE,as well as automatic amino-acid analyzor were drawn to investigate the hydrolyzing ability of MCRC-Ce( Ⅲ) toward BSA. The results indicated that after 24 hours,BSA can be hydrolyzed up to 88%at physiological pH,60℃,where protein was hydrolyzed into smaller peptides and amino acids,and the pseudo-first-order kinetic constant( Kobs) was 7.05 ×10- 2mol·L- 1·h- 1. MCRC-Ce( Ⅲ) has a strong binding affinity with metal ions,which could be reused for 6 times while the catalytic ability maintains 80% of the original,indicating that MCRCCe( Ⅲ) has a broad application prospect.
