前列腺素受体基因多态性与曲伏前列腺素降眼压疗效的相关性

【摘要】 目的观察前列腺素受体基因(PTGFR)多态性与曲伏前列腺素降眼压疗效的相关性。方法纳入50例原发性开角型青光眼或高眼压症患者,用PCR产物直接测序法对PTGFR基因的第2,3,4外显子测序及位于启动子的4个SNPs位点rs3753380、rs3766355、rs1073610、rs1073611、位于第1内含子的rs3766355等位点进行基因分型;分析其与曲伏前列腺素降眼压疗效的关系。结果PTGFR基因的第2,3,4外显子未发现突变位点,2例在rs1073610G/A、rs1073611A/G发现突变位点。rs3766355C/A位点基因多态性与基线眼压相关(P<0.05);在高眼压症患者中rs3753380T/C位点、rs3766355A/C位点基因多态性与曲伏前列腺素降眼压疗效相关。结论 PTGFR基因rs373380位点、rs3766355位点基因多态性与高眼压症患者使用曲伏前列腺素的降眼压疗效相关。

【Abstract】 Objective To study the association between genetic polymorphisms of prostaglandin receptor( PTGFR) gene and the effect of travoprost. Methods A pharmacogenetic analysis was performed on 50 Chinese patients with primary open- angle glaucoma or ocular hypertension who were treated with travoprost. The second,third,forth exon and three SNPs( rs3753380,rs3766355,rs1073610,rs1073611) in promoter,one SNP( rs3766355) in intron of PTGFR were chosen for genotypic analysis using direct DNA sequencing method. Results Mutation site was not found in the second,third,forth exon of PTGFR. Mutation sites of rs1073610G / A and rs1073611A / G were found in two patients. Rs3766355C / A gene polymorphism had a correlation with the baseline level of intraocular pressure( P < 0. 05). Rs3753380T / A and rs3766355A /C gene polymorphism had a correlation with effects of travoprost in patients of ocular hypertension. Conclusion SNPs of rs3753380and rs3766355 in PTGFR are associated with the effect of travoprost on patients with ocular hypertension.
