维生素K2对SCA3/MJD转基因果蝇的神经保护作用研究

【摘要】 目的:探讨维生素K2对SCA3/MJD的神经保护作用。方法:构建Gmr-GAL4/UAS系统和Nrv2-GAL4/UAS系统两种不同表达部位的SCA3/MJD转基因果蝇模型。并分别用1.00,0.10和0.01mM的维生素K2干预。结果:0.10mM的维生素K2明显抑制了SCA3/MJD转基因果蝇眼睛视网膜光感受神经元变性,降低果蝇翅膀异常率,改善果蝇运动能力,0.01mM的维生素K2作用较0.10mM的弱,1.00mM的维生素K2反而加重SCA3/MJD转基因果蝇眼睛视网膜光感受神经元变性,增加果蝇翅膀异常率,降低果蝇运动能力。结论:维生素K2对SCA3/MJD转基因果蝇的神经保护作用与维生素K2剂量有关,过低作用弱,而过高损伤神经元。

【Abstract】 Objective:To investigate the neuroprotective effect of vitamin K2on SCA3/MJD.Methods:To construct Gmr-GAL4/UAS and Nrv2-GAL4/UAS systems,which expressed at different parts in SCA3/MJD transgenic Drosophila models.SCA3/MJD transgenic Drosophila models were interfered with 1.00,0.10and 0.01mM.Vitamin K2.Results:0.10mM vitamin K2significantly inhibited SCA3/MJD transgenic Drosophila eye retinal photoreception neuronal degeneration,reduced the abnormal rate of Drosophila wings,improved exercise capacity,the effect of 0.01mM vitamin K2was weaker than that of 0.10mM vitamin K2,1.00mM of vitamin K2increased SCA3/MJD transgenic Drosophila eye retinal photoreception neuronal degeneration,increased anomaly rate of Drosophila wings,reduced the locomotor ability.Conclusion:The neuralprotective effect of vitamin K2on SCA3/MJD transgenic Drosophila is associated with vitamin K2dose,too low,weak role,and too much,damage to neurons.
