【摘要】 目的探讨膀胱尿路上皮癌组织中IGF-1R和MMP-2蛋白的表达及临床意义。方法收集武汉大学人民医院病理科2000-2006年有完整临床和病理资料的膀胱尿路上皮癌存档蜡块50例和5例癌旁组织,采用免疫组织化学S-P法检测50例膀胱尿路上皮癌和5例癌旁组织中IGF-1R和MMP-2蛋白的表达水平,并分析他们与临床病理特征的相关性。采用HPIAS-1000高清晰度彩色病理图文报告管理系统,对IGF-1R和MMP-2蛋白的表达进行定量分析,并用SPSS13.0软件对各组免疫组织化学反应阳性颗粒的平均光密度、阳性面积率做单因素方差分析和SNK(q)检验。结果 (1)IGF-1R和MMP-2蛋白在膀胱尿路上皮癌中呈高表达,癌旁组织中呈低表达。膀胱尿路上皮癌与癌旁组织相比,差异有显著性(P<0.05);(2)IGF-1R和MMP-2蛋白的表达与膀胱尿路上皮癌的肿瘤大小、淋巴结转移和临床分期有关(P<0.05);(3)IGF-1R和MMP-2蛋白的表达与膀胱尿路上皮癌患者的病理分型、患者年龄、性别无关(P>0.05)。结论 IGF-1R和MMP-2与膀胱尿路上皮癌的发生、发展和转移过程密切相关,对临床上进一步治疗提供参考依据。更多还原

【Abstract】 Objective To investigate the relationship between the expressions of insulin-like growth factor-1receptor(IGF-1R)and matrix metalloproteinase-2(MMP-2)protein in bladder urothelial cancer.Methods Parafflin-embeded specimens of 50 cases of bladder urothelial cancer,and 5cases of normal tissues near the cancer,which were all filed in full pathological and clinical data,were collected from the pathology department of the People′s Hospital of Wuhan University between 2000 and 2006.The expression of IGF-1Rand MMP-2Protein were detected by immunohistochemical S-P method;then their relevance with clinicopathological features was analysed.Quantitative analysis of IGF-1Rand MMP-2protein was conducted by high-definition pathological graphics context report system(HPIAS-1000).One-way analysis of variance and SNK(q)tests were used to analyse the mean density and the positive area rate of the immunohistochemical results.Results(1)The expressions of both IGF-1R and MMP-2 Protein in bladder urothelial cancer were significantly higher than that in normal tissues(P<0.05);(2)The expressions of IGF-1Rand MMP-2Protein were significantly associated with bladder urothelial cancer size,lymph node metastasis,Fuhrman classification and Robson staging(P<0.05);(3)The expressions of IGF-1Rand MMP-2protein had no correlation with the pathologic types,patient age,gender(P>0.05).Conclusion High expression of IGF-1Rand MMP-2protein are significantly correlated with growth,invasion and metastasis of bladder urothelial cancer,and therefore could be taken as a vital index to judge the feature of bladder urothelial cancer and reference of further therapy.更多还原
