基于PEG-DOPA的可降解水凝胶的制备及性质
Bio-Inspired Degradable Hydrogel Based on PEG and DOPA
【摘要】 多巴(DOPA)作为一种生物配体,由于其独特的生物粘附性在近来的研究中受到广泛关注。本研究中将DOPA分子和二硫键修饰于四臂聚乙二醇分子(T-PEG)的末端,利用高碘酸钠(NaIO4)实现其快速交联,得到了水凝胶。研究表明,该水凝胶在不同浓度的谷胱甘肽(GSH)作用下可实现在不同时间的降解,DOPA分子的引入显著提高了该水凝胶的机械强度。3D细胞培养及毒性测试表明,该水凝胶对骨髓间质干细胞(BMSCs)的生长有较好的促进作用。
【Abstract】 Four armed polyethylene glycol(T-PEG)was modified with DOPA and disulfide bonds at the chain end to afford the gelator of PEG-DOPA.PEG-DOPA could form hydrogel rapidly with sodium periodate(NaIO4)in the system.It was found that the hydrogel degraded in the presence of glutathione solution(GSH),and the degradation time could be adjusted by using different concentrations of GSH.The mechanical strength was obviously enhanced with DOPA in the structure of gelator,and the mechanical modulus of hydrogels increased with increasing the concentration of PEG-DOPA.BMSCs cells were cultured in the hydrogels that showed no cytotoxicity.
