维甲酸诱导的骨髓间充质干细胞移植后向少突胶质体细胞分化及Foxp3表达的研究

【摘要】 目的探讨维甲酸诱导的骨髓间充质干细胞(MSCs)移植到脱髓鞘脊髓内向少突胶质细胞分化及Foxp3表达。方法用脊髓内注射溴化乙锭制备脱髓鞘脊髓大鼠模型,移植维甲酸体外诱导MSCs,30 d后灌注取材,采用免疫荧光双标染色检测少突胶质细胞标志物APC和Treg细胞标志物Foxp3表达。结果维甲酸诱导MSCs移植到脱髓鞘脊髓30d后,约有(15.0±4.5)%分化为少突胶质细胞,FoxP3阳性率为(86.2±12.9)%。结论维甲酸诱导的MSC移植到脱髓鞘脊髓可向少突胶质细胞分化,并表达FoxP3。
【Abstract】 Objective To investigate the oligodendrocyte differentiation and Foxp3 expression of retinoic acid(RA)-induced bone marrow mesenchymal stem cells(MSCs) after transplanted into the spinal demyelination. Methods The spinal demyelination rats were established by spinal injection of ethidium bromide and then transplanted with RA-induced MSCs. Expression of APC(oligodendrocyte marker) and Foxp3(Treg cell marker) in spinal cord was detected by double immunofluorescence staining after 30 days. Results The differentiation rate of oligodendrocytes and FoxP3 positive rate of RAinduced MSCs were(15.0±4.5)% and(86.2±12.9)%, respectively. Conclusion RA-induced MSCs can differentiate into oligodendrocytes and express FoxP3 after transplanted into the spinal demyelination.
