【摘要】 目的构建小鼠Th17细胞体外诱导分化模型。方法提取小鼠脾淋巴细胞,采用免疫磁珠纯化++CD4 CD62L T细胞(原始T细胞),分对照组及Th17组。Th17组采用CD3、CD28、IFN-、IL-4抗体及IL-6、TGF-1、IL-23培养72 h,采用Real-time PCR检测RORt和IL-17F mRNA表达。结果 Th17组T细胞RORt、IL-17F mRNA表达明显高于对照组(P<0.01)。结论构建Th17细胞体外诱导分化模型可用于T细胞分化研究。更多还原

【Abstract】 Objective To construct an in vitro Th17 cell differentiation model in mice. Methods Mouse spleen + +lymphocyt-es were purified by using CD4 CD62L T cell isolation kit II, and then divided into control and Th17 groups. T cells were cultured with anti-mouse CD3, CD28, IFN-, and IL-4 antibodies, and IL-6, TGF-a1, and IL-23 for 72 hours. After harvesting, RORt and IL-17F mRNA expression were detected by real-time PCR. Results RORt and IL-17 F mRNA expression was upregulated in Th17 group compared with control group(P<0.01). Conclusion Construction of the in vitro Th17 cell differentiation model can be feasible in the study of T cell differentiation.更多还原
