中国龙虾食物过敏反应中动物肺组织的变化研究

【摘要】 将40只雄性Balb/c小鼠分为处理组〔中国龙虾蛋白组、卵清蛋白(OVA)组〕、阴性对照Coca’s液组和空白对照组,用中国龙虾蛋白或OVA混合氢氧化铝佐剂后进行腹腔注射致敏并通过灌胃激发建立食物过敏反应小鼠。ELISA法测定致敏激发后的小鼠血清中IgE与组胺水平,收集支气管肺泡灌洗液并进行细胞计数,同时观察肺的组织学改变。灌胃激发后处理组小鼠出现身体抖动、呼吸急促等症状,处理组小鼠血清IgE、组胺与Coca’s液组或正常对照组相比显著升高;中国龙虾组和卵清蛋白组小鼠肺泡灌洗液中白细胞总数分别为37.5±3.4和41.2±4.1、嗜酸性粒细胞数分别为18±6和15±4,与正常对照组或Coca’s液组相比皆显著提高;处理组肺间质和肺泡内可见以淋巴细胞、嗜酸性粒细胞和巨噬细胞为主炎细胞浸润,终末细支气管上皮细胞部分断裂及脱落,管壁增厚。处理结果表明:食物过敏反应中肺也是一个受损伤的器官,肺组织淋巴细胞、嗜酸性粒细胞等炎症细胞浸润增加可能是导致哮喘发生的免疫学基础,提示食物过敏和哮喘发生之间具有一定的相关性。

【Abstract】 Forty male Balb/c mice were divided into three groups: experimental groups(Panulirus stimpsoni protein subgroup and ovalbumin subgroup), Coca’s liquid negative control group and blank group. Immunized by intraperitoneal injection of P. stimpsoni proteins or ovalbumin with aluminum hydroxide, challenged by lavage with the same allergen, the food allergy mice were established. The IgE and histamine in serum were measured by ELISA. Numbers of inflammatory cells and eosinophils(EOS) in bronchoalveolar lavage fluid(BALF) were counted. Lung tissues were observed for histological alteration in light microscope. Symptoms(shivering, tachypnea, et al.) were presented in experimental group mice after challenged with allergens. Compared with Coca’s liquid group or blank group,the serum IgE and histamine level of experimental group increased significantly. Numbers of inflammatory cells of BALF in P. stimpsoni protein subgroup and ovalbumin subgroup were 37.5±3.4, 41.2±4.1 respectively, and numbers of EOS were 18±6, 15±4, respectively. Infiltration of inflammatory cells(lymphocytes, eosinophils, macrophages, et al.) into mesenchymal and alveolus, thickening of the wall of terminal bronchiole, and epithelial cells break and shedding, were observed evidently in the pulmonary tissues of the mice in P. stimpsoni protein subgroup and ovalbumin subgroup. The results inclicate that lung is an organ affected by food allergy. Infiltration of inflammatory cells(lymphocytes, eosinophils, macrophages, et al.) into mesenchymal and alveolus of lung may be the immunological basis of asthma. There is certain correlation between food allergy and asthma.
