DNA甲基转移酶1与3B在子宫内膜异位症血与腹腔液的表达及相关性研究

【摘要】 目的测定不同期别子宫内膜异位症(endometriosis,EMS)患者血液及腹腔液中DNA甲基转移酶(DNA methyltransferase,DNMT)1及DNMT 3B的表达水平,探讨EMS DNMT 1及DNMT 3B的表达水平与发病的关系。方法采用酶联免疫吸附方法 (ELISA)测定56例EMS患者(35例Ⅰ~Ⅱ期、21例Ⅲ~Ⅳ期)及25例非内异症对照者血及腹腔液中DNMT 1、DNMT 3B的水平,分析其改变与疾病的关系及其二者在两样本中的相关关系。结果 EMS组血中DNMT1、3B及腹腔液DNMT 3B水平明显高于对照组(P<0.05)。血DNMT1、DNMT3B的表达水平在EMSⅢ~Ⅳ期高于Ⅰ~Ⅱ期与对照组,Ⅰ~Ⅱ期DNMT 3B亦较对照组高,差异有统计学意义(P<0.05);腹腔液中DNMT 1、3B的表达水平在EMSⅠ~Ⅱ期高于Ⅲ~Ⅳ期与对照组,其中DNMT 3B在三组之间差异有统计学意义(P<0.05),而DNMT 1的组间差异无统计学意义(P>0.05)。增殖期腹腔液中DNMT 1水平高于分泌期(P<0.05)。血中DNMT 1与DNMT 3B表达呈现正相关关系,腹腔液中的二者亦为正相关(r血清=0.252,P血清=0.023;r腹腔液=0.379,P腹腔液=0.000);DNMT 3B水平在血与腹腔液中呈正相关,DNMT 1在两样本中无相关性。结论 EMS患者DNMT1及DNMT3B表达水平的升高,及不同期别EMS血与腹腔局部DNMT的差异性变化,提示甲基化改变在EMS发生发展中起重要作用。

【Abstract】 Objective To study the relationship between DNA methyltransferase(DNMT)1,DNMT 3B and the development of endometriosis(EMS)by detecting the level of DNMT1 and DNMT3Bin both blood and peritoneal fluid in EMS.Methods The levels of DNMT1 and DNMT 3Bin 56 EMS patients(35withⅠ~Ⅱstages and 21 with Ⅲ~Ⅳ stages)and 25 control patients were measured by enzyme linked immunoabsorbent assay(ELISA).Results The blood level of DNMT1,DNMT 3Band the peritoneal level of DNMT3 Bin EMS group were higher than that in control group(P <0.05).The blood levels of DNMT1 and 3Bin patients withⅢ~Ⅳstages were higher than that in patients withⅠ~Ⅱstages and controls,and DNMT3BinⅠ~Ⅱstages was also higher than controls(P<0.05).The peritoneal fluid levels of DNMT1 and 3Bin patients withⅠ~Ⅱstages were higher than that in patients withⅢ~Ⅳstages and controls,and DNMT3 Blevel among the three groups had statistically significance(P<0.05),while DNMT1 level in the three groups had no obvious difference(P>0.05).Peritoneal fluid DNMT1 level in proliferative phrase was higher than that in secretory phase(P=0.007).DNMT1 and DNMT3Blevel had positive correlation in blood,as well as in peritoneal fluid(rserum=0.252,Pserum=0.023;rPF=0.379,PPF=0.000).DNMT3 Blevel in blood and peritoneal fluid had positive correlation,while this relationship was not found in terms of DNMT1.Conclusion The levels of DNMT1 and DNMT3Bin EMS were increased,and different stages of EMS had different variation both in blood and in local peritoneal cavity,which indicate that DNA methylation may play an important role in the genesis and development of EMS.
