沼泽红假单胞菌发酵培养条件优化研究
Studyonoptimization of fermentation conditions for Rhodopseudomonaspalustris
【摘要】 试验确定了由实验室保藏的沼泽红假单胞菌R1菌株适宜的发酵培养条件,为其作为饲料添加剂生产应用打下基础。采用正交试验L16(45)的设计对发酵培养基配方进行优化,采用单因素随机试验对培养条件进行优化。试验确定的发酵培养基配方为:甘油1.0 g、氯化铵1.0 g、磷酸氢二钾0.3 g、微量元素溶液0.05 ml、维生素溶液0.15 ml、蒸馏水1 000 ml;最佳培养条件为:初始pH值7.0、培养温度30℃、接种量6%、装液量为液体培养基50 ml/250 ml三角瓶,40 W白炽灯下距离20 cm处照射静置培养。在此条件下,发酵培养7 d,沼泽红假单胞菌R1菌数OD660值可达2.066,沼泽红假单胞菌R1菌数可达5.6×108个/ml。
【Abstract】 The optimization of fermention conditions for Rhodopseudomonas palustris R1 in laboratorywas made, so as to provide the basis for its production and application as feed additives. Composi-tions of medium and cultivating conditions were optimizated by L16(45) orthogonal experiment and sin-gle factor test. The optimum medium compositions were: glycerol 1.0 g, NH4 Cl 1.0 g, K2HPO4 0.3 g,trace elements solution 0.05 ml, vitamins solution 0.15 ml, water 1 000 ml; The optimum cultivatingconditions were: the initial pH 7.0, temperature 30 ℃, the volume of inoculum 6%, the volume of medi-um 50 ml/250 ml fermention bottle, fermenion cultivation under 40 W incandescent lamp from 20 cm.In this way, OD660 was up to 2.066 and the number of Rhodopseudomonas palustris R1 reached 5.6×108cells/ml after 7 days cultivation.
