白细胞介素-1β在过敏性鼻炎小鼠的表达及其对鼻上皮细胞肿瘤坏死因子α诱导蛋白2的影响

【摘要】 目的探讨白细胞介素1β(IL-1β)在过敏性鼻炎(allergic rhinitis,AR)小鼠鼻黏膜的表达及其体外对鼻上皮细胞肿瘤坏死因子α诱导蛋白2(tumor necrosis factor-α-inducible protein-2,TNFAIP2)表达的影响。方法建立AR小鼠模型,检测鼻黏膜IL-1β基因的表达;培养小鼠鼻原代上皮细胞,用不同剂量IL-1β刺激,检测上皮细胞中TNFAIP2基因和蛋白的表达。结果 AR小鼠鼻黏膜IL-1β基因表达相对值为8.7±1.74,显著高于对照组1.21±0.26(P=0.0017);小鼠鼻上皮细胞经25、50 ng/ml IL-1β刺激后TNFAIP2基因表达的相对值分别为15.83±1.65、26.50±4.25,均显著高于对照组2.31±0.78(P<0.0001,P=0.0002),且50 ng/ml刺激组显著高于25 ng/ml刺激组(P=0.04);TNFAIP2蛋白表达呈相同趋势。结论 AR小鼠鼻黏膜IL-1β表达增强,IL-1β体外能够诱导鼻上皮细胞表达TNFAIP2,提示AR中IL-1β可能通过调节TNFAIP2的表达发挥作用。

【Abstract】 OBJECTIVE To investigate the expression of interleukin-1β(IL-1β) in the nasal mucosa of the allergic rhinitis(AR) mouse and the effect of IL-1β on tumor necrosis factor-α-inducible protein-2(TNFAIP2) expression in primary nasal epithelial cells in vitro. METHODS AR mouse model was developed and IL-1β gene expression in the nasal mucosa was measured. Primary epithelial cells were cultured from mouse nasal tissue and stimulated with different concentration of IL-1β in vitro, the expression of TNFAIP2 gene and protein was measured. RESULTS The relative value of IL-1β gene in the nasal mucosa of AR mouse was 8.7±1.74, significantly higher than that of the control mice(1.21±0.26)(P=0.0017). The relative value of TNFAIP2 gene in mouse epithelial cells stimulated with 25 and 50ng/ml IL-1β(15.83±1.65 and 26.50±4.25 respectively) were both higher than that in cells cultured with medium control(2.31±0.78)(P<0.0001, P=0.0002), and it was higher when stimulated with 50 ng/ml IL-1β than with 25 ng/ml(P=0.04), expression of TNFAIP2 protein had the same trend. CONCLUSION Expression of IL-1β was increased in AR mouse and IL-1β can stimulate the production of TNFAIP2 in nasal epithelial cell in vitro, which indicate that IL-1β may act in AR via up-regulating the expression of TNFAIP2.
