牛支原体分离株全基因组序列测定及初步分析
Complete Genome Sequence and Preliminary Analysis of Mycoplasma bovis Isolated Strain
【摘要】 为全面了解牛支原体NM2012株的基因序列,研究其潜在的致病机制及其复制、转录、翻译过程的调控机制,采用体外大量培养,酚-氯仿法提取牛支原体NM2012株基因组DNA,并送往北京华大基因公司进行全基因组测序注释。结果显示,牛支原体NM2012株基因组大小为993 483bp,GC含量为29.26%,测序组装后共产生3个scaffolds,24个contigs。基因组组分分析后发现,NM2012株的基因组含有885个基因,总长度为834 042bp,基因平均长度为942bp,占基因组全长的83.95%。串联重复序列共79个,总长为9 203bp,占基因组全长的0.926 3%。小卫星序列53个,微卫星序列5个,tRNA 34个,rRNA 4个。牛支原体NM2012株16SrRNA序列与支原体属致牛羊病主要菌株构建进化树进一步证实NM2012株为牛支原体,为后期进一步研究牛支原体提供依据。
【Abstract】 The purpose of this test was to understand the genetic sequence of Mycoplasma bovis NM2012 strain,and study its potential pathogenesis and its regulation mechanisms of replication,transcription and translation.NM2012 strain in vitro and its genomic DNA was extracted by phenol-chloroform extraction method and sent to the Beijing Genomics Institute for whole genome sequence.The result showed that the genome size of Mycoplasma bovis NM2012 strain is 993 483 bp,GC content is 29.26%,a total of three scaffolds and 24 contigs.Genome analysis found that Mycoplasma bovis NM2012 genome contains 885 genes,total length is 834 042 bp,average length is 942 bp,representing 83.95% of the full length genome.Tandem repeats is 79,total length is 9 203 bp,accounting for 0.926 3% of full-length genome.There are 53 small satellite sequences,5microsatellite sequences,34 tRNA and 4rRNA.By constructing phylogenetic tree with 16 SrRNA sequence from sequenced Mycoplasma bovis NM2012 strain and Mycoplasma which mainly infecting cattle and sheep,we further confirmed that NM2012 strain is Mycoplasma bovis.The study provides basis for further research on Mycoplasma bovis.
