感染高原鼠兔的皮蝇蛆线粒体CO1基因序列分析
Sequence Analysis of Mitochondrial Cytochrome Oxidase 1 (CO1) Gene of Hypoderma Larvae Plateau Pika
【摘要】 为分析感染高原鼠兔的皮蝇蛆分子分类地位,通过聚合酶链反应(PCR)对6株感染高原鼠兔的皮蝇蛆的线粒体CO1基因片段进行扩增、克隆和测序,并将其序列与GenBank中的皮蝇科、胃蝇科、狂蝇科的序列进行了比对分析,构建了遗传进化树。结果显示,扩增出的CO1基因片段为688bp,感染高原鼠兔的皮蝇蛆与皮蝇科的皮蝇蛆遗传距离更近,位于一个大的进化树分支上,其基因片段同源性为81.2%~86.6%;与胃蝇科的蝇蛆基因片段同源性为77.8%~80.4%,与羊狂蝇基因片段同源性为79.4%~79.8%。结果表明,试验中的6株感染高原鼠兔的皮蝇蛆之间的基因片段同源性98.1%~100%,为皮蝇科的皮蝇蛆。
【Abstract】 The objectives of this study were to analyze of molecular taxonomic positions of Hypodermalarvae in the plateau pika.The partial sequence of cytochrome oxidase 1gene(CO1)from 6strains of Hypodermalarvae in plateau pika were amplified,cloned,sequenced and compared with Hypodermatidae,Gasterophilidae,Oestridae in GenBank,and phylogenetic analyses were also constructed.The results showed that Hypodermalarvae in the plateau pika and other Hypodermalarvae have a closer genetic relationship,clustered in one large branch of the phylogenetic tree,with 81.2%-86.6%in genetic homology;and the sequence identities is 77.8%-80.4% with Gasterophilidae larvae,and 79.4%-79.8% with Oestrus ovis.The genetic similarity of CO1fragments between 6 Hypodermalarvae in plateau pika is 98.1%-100%,belonging to Hypodermatidae.
