不同来源嗜水气单胞菌溶血素基因检测及序列分析

【摘要】 对分离自发病宽体金线蛭(Whitmania pigra)、中国林蛙(Rana chensinensis)、牙鲆(Paralichthys olivaceus)、草鱼(Ctenopharyngodon idellua)、青鱼(Mylopharyngodon piceus)、鲤(Cyprinus carpio)的42株致病性嗜水气单胞菌(Aeromonas hydrophila)采用PCR方法,进行了溶血素(Ahh)基因的检测,并将代表菌株的Ahh基因片段通过Blast与GenBank中报道的相似性较高的10株嗜水气单胞菌参考菌株进行了核苷酸同源性分析。结果显示,所检不同来源的嗜水气单胞菌均携带Ahh基因;所测代表菌株Ahh基因片段与10株嗜水气单胞菌参考菌株的同源性为94.1%~98.9%;供试不同来源嗜水气单胞菌株间Ahh基因的同源性为97.1%~100%,其中分离自草鱼的HC960715-5与分离自宽体金线蛭的HTQ010505-3之间同源性高达100%,分离自青鱼的HL060811-3与分离自林蛙的HQ070610B-3同源性也高达100%。

【Abstract】 Forty-two pathogenic Aeromonas hydrophila strains isolated from Whitmania pigra,Rana chensinensis,Paralichthys olivaceus,Ctenopharyngodon idellua,Mylopharyngodon piceus and Cyprinus carpio were investigated for the presence of hemolysin( Ahh) gene using PCR method,and the homology of Ahh gene sequences from diverse representative A. hydrophila strains were compared with available sequences in the NCBI GenBank using the soft-ware DNAStar. The results showed that all the detected strains were positive for Ahh gene. The Ahh genes of representative strains which were isolated from different hosts shared 94. 1% ~ 98. 9% nucleotide sequence homology with the reference strains. The Ahh genes of 7 representative strains showed a higher percentage of similarity( 97. 1% ~ 100%). Among them,Ahh gene sequence homologies of representative strains HC960715-5 isolated from C. idellua and HTQ010505-3 isolated from W. pigra, as well as HL060811-3 isolated from M. piceus and HQ070610B-3 isolated from R. chensinensis,were as high as 100%.
