黑豆萌芽中抗氧化活性相关基因表达研究

【摘要】 采用基因芯片技术和GO Term分析研究了黑豆萌芽中抗氧化活性相关基因的表达,并通过RT-PCR进行验证。结果表明:对比0.5和5.0 cm黑豆萌芽的差异表达基因,在芽长0.5 cm的萌芽中发现了10个与抗氧化活性相关的基因显著高表达,他们属于GO:0006979(response to oxidative stress,氧化压力响应):BE823689.1567,GMFL01-02-F14-R381,GMFL01-03-G22-R364,GMFL01-14-M12-R553,GMFL01-51-M23-R265,AW757007.1297,AW761420.1260,BI788389.1501,BQ273202.1332,GMFL01-10-I14-F701,这些基因主要控制合成热休克蛋白和抗氧化酶类,与细胞内的氧化代谢平衡密切相关;RT-PCR结果与基因芯片试验结果一致,证明了基因芯片结果的准确性和可重复性。

【Abstract】 The purpose of this study was to analyze the gene expression of black soybean(Glycine max var) sprouts with high antioxidant activity.Microarray and GO(Gene Ontology) term enrichment analysis were used to research the expressed genes,and the results were confirmed by qRT-PCR.Microarray analysis of expressed genes between sprouts(0.5 and 5.0 cm) showed that the most up-regulated genes in 0.5 cm sprouts belonged to GO:0006979(response to oxidative stress),in which ten genes were significantly highly expressed:BE823689.1567,GMFL01-02-F14-R381,GMFL01-03-G22-R364,GMFL01-14-M12-R553,GMFL01-51-M23-R265,AW757007.1297,AW761420.1260,BI788389.1501,BQ273202.1332 and GMFL01-10-I14-F701.Those genes mainly control the synthesis of heat shock protein and antioxidant enzymes which related to intracellular oxidation metabolic balance.The results of qRT-PCR analysis regarding up-regulated genes were consistent with those of the microarray analysis,which indicated that the results were biologically reproducible and believable.
