LC-UV法测定华中五味子中五味子酯甲、五味子甲素、五味子乙素含量
Simultaneous determination of schisantherin A,deoxyschizandrin andγ-schisandrin in S. sphenanthera Rehd.et Wils.by LC-UV
【摘要】 目的建立同时测定五味子酯甲、五味子甲素、五味子乙素的LC-UV含量测定方法。综合考察皖南山区野生华中五味子的内在品质,为该品种转为栽培品提供理论依据。方法采用LC-UV法,Agilent TC-C18(2)色谱柱(250 mm×4.6 mm I.D,5 um),以甲醇-水为流动相,梯度洗脱,流速为1 mL/min,柱温30℃,检测波长为240 nm,测定五味子酯甲、五味子甲素、五味子乙素。结果皖南山区野生华中五味子中五味子酯甲、五味子甲素、五味子乙素平均含量分别为0.24%、0.46%、1.22%。结论皖南山区野生华中五味子中五味子酯甲平均含量为0.24%,符合《中国药典》2010版的要求:五味子酯甲含量不低于0.20%,可供药用。通过三指标的综合控制,为皖南华中五味子的品质评价和栽培提供理论支持。
【Abstract】 Objective To investigate the determination of the contents of Schisantherin A、Deoxyschizandrin andγ-schizandrin in S.sphenanthera Rehd.etWils.by LC-UV and explore the quality of wild S.sphenanthera Rehd.etWils.in Wannan mountainous area to provide a reliable measure forquality control ofGAP.Methods 1.LC-UVmethodwasestablishedwith a Agilent TC-C18(2)column(5μm,4.6× 250 mm),a mobile phase of Methanol-Water solution a flow rate of 1.0 mL· min- 1 and a detectionwavelengthof 240 nminorderto determine the contentsofSchisantherainA、Deoxyschizandrin andγ-schizandrin.The column temperature was maintained at 30 ℃.Results The average contents of Schisantherain A、Deoxyschizandrin andγ-schizandrin in the decoction of wild S.sphenanthera Rehd.etWils.in Wannan mountainous area were 0.24%、0.46%、1.22% respectively.Conclusion The average contentof SchisantherinA inwild S.sphenanthera Rehd. etWils in Wannan mountainous area was 0.24% which is in accordance with the requirements Chinese Pharmacopoeia(2010 Edition).The contentofSchisantherinAis no less than 0.20%,which can be used formedicine.By the comprehensive control of three indicators,theoretical support for the quality evaluation and cultivation of S.sphenanthera Rehd.et Wils.in southern Anhui can be provided.
