长根金星蕨总黄酮抗氧化活性研究

【摘要】 研究长根金星蕨中总黄酮的抗氧化性。采用70%乙醇提取长根金星蕨中总黄酮,用NaNO2-Al(NO3)3-NaOH分光光度法测定总黄酮含量。用Fenton体系、邻苯三酚体系、普鲁士蓝法研究总黄酮溶液的体外抗氧化活性,用硫代巴比妥酸(TBA)分光光度法研究其对羟自由基引发DNA氧化损伤的抑制作用。样品中总黄酮含量为1.95%。总黄酮浓度为60μg/mL时,对羟自由基的清除率可达30.1%;浓度为67μg/mL时,对超氧阴离子自由基的清除率可达50.2%;浓度为87μg/mL时,对羟自由基引发DNA损伤的抑制率可达95.9%。结果表明长根金星蕨中总黄酮能较为有效清除活性氧自由基,对DNA氧化损伤有显著抑制作用。

【Abstract】 This paper presents a study onthe anti-oxidative activity of total flavonoids in Parathelypteris beddomei. For this purpose,the flavonoids were extracted with 70% ethanol,its content determined by NaNO2-Al( NO3) 3-NaOH spectrophotometry,the anti-oxidative activity of the extraction solution evaluated with Fenton,pyrogallol as well as Prussian blue systems,and the inhibitory effects in Parathelypteris beddomei on demage of DNA chain induced by hydroxyl radical that was observed by thiobarbituric acid spectrphotometry. Experiments demonstrated that the content of the total flavonoids in Parathelypteris beddomei was 1. 95%; when the concentration of total flavonoids was 60μg /mL,it could purge away 30. 1% of hydroxyl free radicals; when the concentration was 67μg /mL,50. 2% of superoxide anion free radical could be cleared up; and at 87μg /mL the DNA damage due to free hydroxyl radicals could decrease by 95. 9%. Therefore,it could be concluded that the total flavonoids in Parathelypteris beddomei can purge away active oxygen efficiently and,consequently,inhibit DNA damages obviously.
