ω-3多不饱和脂肪酸对严重烧伤大鼠肾组织及其炎性反应的影响
Effects of ω-3 polyunsaturated fatty acids on kidney and inflammatory response in burned rats
【摘要】 目的:研究ω-3多不饱和脂肪酸(ω-3 PUFA)对严重烧伤大鼠肾组织及其炎性反应的影响。方法:将72只大鼠随机分成三组,即假烫组(A组)8只、对照组(中长链脂肪酸组,B组)32只和治疗组(ω-3 PUFA组,C组)32只。采用大鼠30%总体表面积(TBSA)Ⅲ度烫伤模型。B组每天按时经尾静脉注射与C组相同热量的中长链脂肪酸,C组经尾静脉注射ω-3 PUFA 2 ml/(kg·d),A组不做任何处置。分别观察A、B、C组在伤后第1、4、7和10天的血清前列腺素E2(PGE2)、白细胞介素-1(IL-1)和肿瘤坏死因子-α(TNF-α)水平。采用普通聚和酶链反应(RT-PCR)检测肾组织TNF-αmRNA的表达。各个时间点均处死8只大鼠。结果:B组和C组大鼠在伤后各时间点血清PGE2、IL-1和TNF-α水平显著高于A组(P<0.01)。C组血清PGE2、IL-1和TNF-α水平低于B组,在伤后第4、7和10天与B组比,差异有显著性统计学意义(P<0.05,P<0.01)。另外,B组和C组大鼠在伤后各时间点肾组织TNF-αmRNA表达较A组明显升高。与B组比较,C组肾组织TNF-αmRNA的表达明显降低。结论:ω-3 PUFA能降低烧伤后大鼠血清PGE2、IL-1和TNF-α水平,减轻烧伤后肾组织TNF-αmRNA的表达,可能对烧伤后肾组织具有保护作用。
【Abstract】 Objective: To study the influence of ω-3 polyunsaturated fatty acids(ω-3 PUFA) on kidney and inflammatory reaction in serious scalded rats. Methods: A total of 72 SD rats with 30%total body surface area(TBSA) third-degree scalded were separated into 3 groups: the phony scald group(group A)8,the control group(middle-long chain fatty acids group,group B) 32 and treatment group(ω-3 PUFA group,group C)32. Rats in group B were injected middle-long chain fatty acids with equivalent calories by tail vein,and rats in group C were injected ω-3 PUFA 2 ml /(kg·d) by tail vein on time for everyday. Rats in Group A had no any disposal. The levels of serum prostaglandin E2(PGE2),Interleukin-1(IL-1) and tumor necrosis factor alpha levels( TNF-α) were examined. Expression of kidney tissue TNF-α mRNA was measured by RT-PCR technology. Results: Serum PGE2、IL-1、TNF-α were significantly higher than pre-injury at each time point after injury in group B & C( P < 0. 01). The levels in group C were lower than in group B,and were significantly different in 4 d,7 d,10 d after injury(P <0.05 or P <0.01). Compared with group B in 4d,7d after injury,the expression of kidney tissue TNF-α mRNA was significantly reduced in group C. Conclusion: ω-3 PUFA can reduce the levels of serum PGE2,IL-1,TNF-α,relieve the expression of kidney tissue TNF-α mRNA,and protect the damage of kidney after burn.
