L-精氨酸对人胃癌细胞株SGC-7901凋亡的调控作用

【摘要】 目的:研究L-精氨酸(L-Arg)对人胃癌细胞株SGC-7901凋亡的调控作用。方法:以胃癌细胞株SGC-7901为研究模型,采用流式细胞术和FITC-AnnexlnV/PI双染色检测L-Arg调控细胞凋亡的作用。借助western blot技术检测细胞凋亡分子的表达,利用RT-PCR方法检测等分子生物学技术对L-Arg胃癌细胞凋亡的调控作用进行研究。结果:L-Arg明显抑制抗凋亡蛋白Bcl-2和survivin的表达,明显上调p53蛋白的表达。结论:L-Arg通过下调抗凋亡蛋白Bcl-2和survivin的表达,上调促凋亡蛋白p53的表达,发挥对人胃癌细胞株SGC-7901凋亡的调控作用。

【Abstract】 Objective: The aim of this study was to explore the regulation of L-arginine on apoptosis in human gastric cancer cell line SGC-7901. Methods: This study took human gastric cancer cell line SGC-7901 as the tumor model. Flow cytometry and FITC-Annexin-V /PI methods were used to detect the apoptosis effect of L-arginine. Western blot was applied to detect the expression of apoptosis signaling molecules. RT-PCR method was performed to explore the regulating effect of L-arginine on apoptosis in human gastric cancer cell. Results: L-arginine could significantly reduce the expression of anti-apoptotic protein Bcl-2 and survivin and raise the expression of pro-apoptotic protein p53. Conclusion:L-arginine regulates apoptosis in human gastric cancer cell by reducing the expression of anti-apoptotic protein Bcl-2 and survivin and raising the expression of pro-apoptotic protein p53.
