二氧化钛/氧化锌复合薄膜的制备及生物相容性表征
Preparation of TiO2/ZnO Composite Film and Characterization of Biocompatibility
【摘要】 采用溶胶-凝胶法,制备了TiO2/ZnO微/纳结构复合薄膜。利用扫描电子显微镜(SEM)观察微/纳结构复合薄膜的形貌,采用X射线光电子能谱(XPS)分析复合薄膜元素的化合价及表面化学成分,利用紫外-可见分光光度计(UV-vis)分析复合薄膜的紫外吸收光谱,并对样品的表面润湿性及其光响应性进行分析,用CCK-8法表征了所制备薄膜对细胞的粘附和增殖性能。结果表明:制备的TiO2/ZnO复合薄膜表面均匀;Ti元素主要以Ti4+形式存在,Zn元素主要以Zn2+形式存在;6种薄膜的禁带宽度相近,吸收边没有表现出特别大的红移或蓝移;与水的接触角测试表明,所制备的薄膜比较利于细胞的粘附和增殖;复合薄膜生物相容性优良,其中TiO2/ZnO(0.14 mol/L)组对成骨细胞的增殖有促进作用,这可能是由于少量锌离子的缓释对成骨细胞的粘附和增殖所致。
【Abstract】 By a sol-gel method, TiO2/ZnO micro-nano structure composite films were prepared. Scanning electron microscope(SEM) was used to observe the morphology of micro-nano structure composite films, and X-ray photoelectron spectroscopy(XPS) was used to analyze the chemical composition. The ultraviolet absorption spectra of the films and surface wettabilities were also characterized with Ultraviolet-visible spectra(UV-vis) and goniometer. CCK-8 method was used to characterize the adhesion and proliferation of cells. The results show that the TiO2/ ZnO composite films are coated on the quartz substrate successfully. Ti mainly exists in the form of Ti4+ and Zn mainly exists in the form of Zn2+. The band gaps of different films are close and the absorption edges do not show obvious red shift or blue shift; Composite films have excellent biocompatibility. Among all the films, the TiO2/ZnO(0.14 mol/L) group is good for proliferation of osteoblast. That is ascribed to slow-release of zinc ions and Zn ion in low concentration is beneficial to the adhesion and proliferation of osteoblast cells.
