离子凝胶法制备EGF-透明质酸-壳聚糖纳米微球
Preparation of EGF-Hyaluronic Acid-Chitosan Nanoparticles by Ionotropic Gelation Technique
【摘要】 以透明质酸、壳聚糖为原料,采用离子凝胶法制备载有表皮生长因子(EGF)的纳米微球。通过正交试验设计研究了壳聚糖与透明质酸质量比、三聚磷酸钠用量、CS溶液酸碱度、反应时间等因素对微球形貌、粒径的大小及其分布、Zeta电位值、包封率、释放度等参数的影响。结果表明,纳米微球最佳的制备工艺为壳聚糖与透明质酸质量比3∶1,三聚磷酸钠用量1.5mg,CS醋酸溶液pH值4.0,反应时间3h。
【Abstract】 The nanoparticles loaded with epidermal growth factor(EGF)were prepared from hyaluronic acid(HA)and chitosan(CS)by ionotropic gelation technique.The influences of the factors including the mass ratio of CS to HA,the amount of sodium triphosphate(TPP),the pH of CS solution and reaction time on the properties of the microsphere such as the morphology,nanoparticle size and its distribution,Zeta value,the entrapment efficiency and release rate of EGF were researched by orthogonal experiment.The results showed that the optimum preparation technology of nanoparticles is as follows:3∶1mass ratio of CS to HA,the amount of TPP for 1.5mg,the pH of CS solution for 4.0,and reaction time for 3h.
