阿拉伯茶染色体核型分析

【摘要】 以源自埃塞俄比亚的阿拉伯茶(Catha eduli)幼嫩芽尖为材料,采用常规的去壁低渗法,研究了紫芽和绿芽2个品种阿拉伯茶的染色体,并与本土生长的同科植物冬青卫茅(Euonymus japonicas)的染色体及核型做了比较分析。结果显示,阿拉伯茶体细胞染色体数目为32条,绿芽种阿拉伯茶染色体相对长度组成为2n=32=6L+8M2+8M1+10S;核型公式为K(2n)=32=11m(2SAT)+5sm;核型不对称系数(AS.K%)为61.48%;核型分类标准,属于"2B"型。紫芽种阿拉伯茶染色体相对长度组成为2n=32=4L+3M2+3M1+6S;核型公式为K(2n)=32=11m(2SAT)+5sm;核型不对称系数(AS.K%)为60.844%;核型分类标准,属于"2B"型。冬青卫茅体细胞染色体数目也为2n=2x=32;染色体相对长度组成为2n=32=3L+3M2+7M1+3S;核型公式为K(2n)=32=10m(2SAT)+6sm;核型不对称系数(AS.K%)为63.05%。

【Abstract】 The morphology and karyotype of chromosome of the Catha edulis were studied using the regular wall degradation and hypotonic method with the bud-tip from two different varieties of Catha edulis,and compared with chromosome of the indigenous Euonymus japonicas L.The results showed that green bud Catha edulis normal diploid2n=32,6L+8M2+8M1+10S,karyotype formula was K(2n)=32 =11m(2SAT)+5sm,which belongs to"2B"type,the AS.K% was 61.48%.Purple bud Catha edulis normal diploid 2n=32,4L+3M2+3M1+6S,karyotype formula was K(2n)=32 =11m(2SAT)+5sm,which belongs to"2B"type,the AS.K% was 60.844%.Euonymus japonicas L.normal diploid 2n=32,the relative length was 2n=32=3L +3M2+7M1+3S,karyotype formula was K(2n)=32=10m(2SAT)+6sm,AS.K% was 63.05%。
