探析接近临床剂量的甘遂半夏汤加减甘遂甘草对腹水模型大鼠肾脏及免疫系统的影响

【摘要】 目的:观察甘遂半夏汤加减甘遂甘草反药组合对腹水模型大鼠肾脏及免疫系统的影响,探讨药效或毒性反应是否与反药组合相关。方法:将Wistar大鼠按体质量随机分为空白组、模型组、阳性药组、甘遂半夏汤全方组(低剂量全方组)、低剂量全方去醋甘遂组、低剂量全方去炙甘草组和低剂量全方去草遂组,除正常喂养外,每天上午各给药组灌胃给药,空白组和模型组灌胃蒸馏水,给药9d后,所有大鼠禁食不禁水12h,取血,离心,检测肾脏及免疫系统相关指标。结果:与空白组比较,模型组CD3、CD4相对计数降低(P<0.05,P<0.01),Ig A、Ig G、Ig M、尿β2-微球蛋白(β2-MG)、尿表皮生长因子(EGF)含量升高(P<0.05,P<0.01);与模型组比较,各给药组对所检测的免疫系统及肾脏相关指标具有一定的改善作用,其中以低剂量全方组的作用较好。结论:低剂量全方组和低剂量全方去掉方中一味反药或两味反药组在腹水模型大鼠细胞免疫、体液免疫和肾脏系统方面未显示出毒性,同时低剂量全方组具有一定的调节腹水模型大鼠细胞免疫、体液免疫和保护肾脏的作用,并且其疗效优于低剂量全方去掉其中一味反药或两味反药组。

【Abstract】 Objective: Observation of the effects of Gansui Banxia Decoction modified Gansui and Gancao anti-drug combination at preferred dose close to clinical application on kidney and immune system functions in ascites rats mode, to explore whether the efficacy or toxicity associated with the anti-drug combination. Methods: Wistar rats were randomly divided into blank group, model group, furosemide group, Gansui Banxia Decoction group, Gansui Banxia Decoction without Cugansui group, Gansui Banxia Decoction without Zhigancao group, Gansui Banxia Decoction without Zhigancao nor Cugansui group. The drug groups were respectively received the drugs by gavage, while the blank group and model group were received distilled water. After gavage for 9 days, all the rats were fasted for 12 hours to collect blood. The relevant indicators of kidney and immune system function were detected. Results: Compared with the blank group, the counts of CD3 and CD4 in the model group were both lower(P<0.05, P<0.01), and the contents of Ig A, Ig G, Ig M, β2-MG and EGF in model group were all higher(P<0.05, P<0.01). Compared with the model group, the relevant indicators of kidney and immune system function in all drug groups were improved, especially in the low dosage Gansui Banxia Decoction group. Conclusion: There is no toxicity in low dosage of Gansui Banxia Decoction and low dosage of Gansui Banxia Decoction minus one or two drugs of anti-drug combination in rats cellular immunity, humoral immunity and kidney function. The low dosage of Gansui Banxia decoction can regulate the rats cellular immunity, humoral immunity and protect the rat kidney function, which is superior to the low dosage of Gansui Banxia Decoction minus one or two drugs of anti-drug combination.
