健脾养正消癥方对环磷酰胺损伤小鼠骨髓细胞的保护作用

【摘要】 目的:探讨健脾养正消癥方对环磷酰胺(CTX)诱导化疗损伤小鼠骨髓的保护作用。方法:通过血细胞分析、骨髓有核细胞计数、骨髓有核细胞的DNA含量测定和骨髓切片组织学变化,检测健脾养正消癥方对化疗损伤小鼠的骨髓保护作用,并通过体外双层琼脂培养法测定中药联合组对化疗损伤小鼠骨髓细胞粒-巨噬细胞集落(CFU-Gm)形成的影响。结果:中药联合组小鼠血细胞、骨髓有核细胞数量比CTX组明显增加(P<0.05),与空白组无明显差异;中药联合组骨髓有核细胞中G0/G1期细胞的百分率较CTX组明显提高,细胞的凋亡率下降(P<0.05);骨髓组织学切片显示,中药联合组对骨髓有明显的促恢复作用。骨髓集落培养显示,中药联合组有显著的增加骨髓集落形成的作用(P<0.05)。结论:健脾养正消癥方对CTX引起的小鼠骨髓细胞的损伤有明显的保护作用。

【Abstract】 Objective: To discuss the protective effects of Jianpi Yangzheng Xiaozheng Formula(JF) on bone marrow cell in mice injured by cyclophosphamide(CTX). Methods: The protective effect of JF on bone marrow cell in mice injured by CXT were measured through blood cell analysis, bone marrow nucleated count, determination of bone marrow nucleated cell DNA content and the histology changes of bone marrow biopsy. The influence of JF on the colony-forming of granulocytes and macrophages(CFU-Gm) of mouse injured by CXT was tested by double-layer agar culture method in vitro. Results: Compared with CXT group, the amount of blood cells and nucleated cells of marrow increased significantly(P<0.05). Compared with control group, there was no significant difference. Compared with CXT group, the proportion of G0/G1 phase cells in cells of marrow was increased, and the apoptosis rate of cells decreased(P<0.05). The bone marrow biopsy showed that JF had an obvious effect on promoting recovery of bone marrow. The result of bone marrow colony culture displayed that JF had a significant role of increasing the colony-forming of bone marrow(P<0.05). Conclusion: JF shows a significant protective role on mice bone marrow cells injured by CXT.
