薯蓣皂苷抑制脂多糖刺激的大鼠系膜细胞增殖及对Wnt通路的影响
Dioscin ameliorates lipopolysaccharide-stimulated rat mesangial cell proliferation through suppressing Wnt pathway and TGF-β1 expression
【摘要】 目的:探讨薯蓣皂苷干预脂多糖刺激的大鼠系膜细胞(MC)增殖及Wnt通路的作用机制。方法:体外培养大鼠MC(HBZY-1),设对照组、脂多糖(LPS)刺激组、LPS+薯蓣皂苷(安全浓度通过检测LDH释放率得到:0.1、0.5、1.0μg/mL)组。药物干预MC 48h后,采用MTT法检测各组MC的增殖情况,采用RT-PCR法及Western blot法检测Wnt4、β-catenin、转化生长因子β1(TGF-β1)基因和蛋白在各组MC中的表达情况。结果:薯蓣皂苷可以抑制LPS所致的MC增殖,下调Wnt4、β-catinin和TGF-β1 mRNA和蛋白水平的表达(P<0.01,P<0.05),并且在安全浓度范围内其抑制作用与剂量呈正相关。结论:薯蓣皂苷可能通过抑制Wnt信号通路,下调TGF-β1表达发挥抑制大鼠MC增殖的作用。
【Abstract】 Objective: To investigate the ef ects and mechanism of Dioscin on lipopolysaccharide(LPS) stimulated rat mesangial cells(MC). Methods: MC were divided into control group, LPS stimulated group, LPS+Dioscin(0.1, 0.5, 1.0μg/mL) group, and cultured for 48 hours. MTT assay was used to detect the proliferation of MC. RT-PCR and Western blot methods were applied to detect Wnt4, β-catenin and TGF-β1 expression of the MC. Results: Dioscin could inhibit LPS-induced MC proliferation and the expression levels of Wnt4、β-catinin and TGF-β1(P<0.01, P<0.05), in a dose dependent manner. Conclusion: Dioscin can inhibit MC proliferation, and its mechanism may be related to inhibition of the Wnt signaling pathway and reduction of TGF-β1 expression.
