10-羟基喜树碱纳米微球的制备及表征
Preparation and characterization of 10-hydroxycamptothecin nanoparticles
【摘要】 采用溶剂-反溶剂重结晶法进行了10-羟基喜树碱纳米化的研究。考察了以水为反溶剂,不同溶剂对10-羟基喜树碱纳米化的影响,以及不同溶剂反溶剂体积比和表面活性剂用量对其颗粒形貌和大小的影响。实验结果表明选择二甲基亚砜为溶剂,水为反溶剂的溶剂反溶剂效果最好,体系重结晶后形成的10-羟基喜树碱颗粒形貌为球形,粒径均一,约为95nm,添加聚乙烯吡咯烷酮增加了纳米化10-羟基喜树碱悬浮液的稳定性。XRD、FT-IR分析测试结果表明,纳米化颗粒为结晶型,结晶度比原料药低,但其化学结构和组分都与原料药相同;纳米化10-羟基喜树碱溶出速率远远大于原料药的溶出速率,在第30min时,纳米化10-羟基喜树碱的溶出累积量已近于100%,而原料药的溶出量仅为11.6%。
【Abstract】 10-Hydroxycamptothecin( HCPT) nanodispersions have been prepared by using an anti-solvent precipitation method. The influence of different solvents on the nanodispersion of 10-hydroxycamptothecin were investigated,with water as anti-solvent. DMSO was chosen as solvent to prepared nanosized 10-hydroxycamptothecin. The influence of the key parameters,including the volume ratio of solvent and anti-solvent and the weight of PVP,on the particle morphology,size and size distribution have been explored. The average particle size was about 95 nm.The XRD spectra show that compared with the crude HCPT,the nano HCPT has lower crystallinity,and the FT-IR spectra show that the nano HCPT and crude HCPT have the same chemical structure and composition. The dissolution rate of nano 10-hydroxycamptothecin was significantly faster than that of the raw material. In the first 30 min,the cumulative dissolved amount of the nano 10-hydroxycamptothecin was nearly 100%,while the dissolution of the raw material was only 11. 6%.
