内生解淀粉芽孢杆菌X-278发酵条件的优化

【摘要】 以内生解淀粉芽孢杆菌为试材,采用单因素和正交实验,通过比较发酵液的生物量及其对棉花黄萎病菌的抑菌活性,对内生解淀粉芽孢杆菌(Bacillus amyloliquefaciens)X-278液体发酵条件进行了优化。结果表明:内生解淀粉芽孢杆菌X-278培养基各组分的最佳配比为葡萄糖1.5%、花生饼粉2%、硫酸铵0.5%、碳酸钙0.1%、硫酸镁0.5%、氯化钠0.5%;最佳发酵培养条件为初始pH 7,培养温度34℃,转速180r/min,种龄为24h,装液量150mL/500mL三角瓶,接种量2%,发酵时间为72h。优化后的发酵液中活菌数量达到1.5×1010 CFU/mL,对棉花黄萎病菌的抑菌圈直径为33.4mm。

【Abstract】 Taking Bacillus amyloliquefaciens as test material,using single factor and orthogonal experiments,and through comparing biomass of fermentation broth and bacteriostatic activity against Verticillium dahliae Kleb,the fermentation conditions of endogenous Bacillus amyloliquefaciens X-278 were optimized.The results showed that the final optimization of the medium and conditions:1.5% glucose,2% peanut power,0.5%(NH4) 2 SO4,0.1% CaCO3,0.5% MgSO4,0.5% NaCl,pH 7,culture temperature 34℃,rotation speed 180r/min,seed liquid age was 24h,liquid volume was 150mL/500mL of triangular flask,inoculum size was 2%,incubation time was 72h.After optimizing,the number of viable bacteria was to 1.5×1010 CFU/mL in the fermentation broth,and the diameter of inhabiting circle against Verticillium dahliae was 33.4mm.
