【摘要】 目的探讨HBe Ag阴性和HBe Ag阳性的慢性乙型肝炎(CHB)患者血清IL-8含量与外周单核细胞IL-8 mRNA表达的相关性,并进一步研究HBe Ag阴性的CHB患者血清IL-8含量与血清ALT、AST酶含量关系。方法 94例CHB患者根据HBe Ag阴性/阳性分为HBe Ag阴性的CHB组(47例)和HBe Ag阳性的CHB组(47例),以ELISA方法和逆转录PCR法检测两组患者血清IL-8和外周单核细胞mRNA表达,罗氏生化分析仪检测血清ALT和AST酶含量。结果 ELISA和逆转录PCR分析HBe Ag阴性CHB患者的血清IL-8含量与外周单核细胞IL-8 mRNA表达分别为252.65±62.04、1.50±0.42;HBe Ag阳性CHB患者的血清IL-8含量与外周单核细胞IL-8 mRNA表达分别为241.83±49.88、1.28±0.36。HBe Ag阴性的CHB患者血清IL-8含量与外周单核细胞IL-8 mRNA表达具有较好的相关性(r=0.655),而HBe Ag阳性的CHB患者的血清IL-8含量与外周单核细胞IL-8 mRNA表达相关性较差(r=0.276)。其中,HBe Ag阴性的CHB患者血清IL-8含量与血清ALT具酶含量有较好的相关性(r=0.489,P<0.05)。结论 IL-8在HBe Ag阴性和HBe Ag阳性的CHB患者的表达存在一定差异,并且IL-8与部分CHB患者肝功能恶化相关,提示IL-8可能参与慢性乙型肝炎进展过程。更多还原

【Abstract】 Objective To discuss the correlation between the content of serum interleukin 8( IL-8) and the expression of peripheral mononuclear cells IL-8 mRNA in chronic hepatitis B( CHB) patients with HBe Ag negative and HBe Ag positive,and to conduct further research on the relationship between the content of serum IL-8 and serum ALT,AST enzyme. Methods Ninety-four cases of chronic hepatitis b patients were divided into HBe Ag negative CHB group and HBe Ag positive CHB group according to the HBe Ag negative / positive,the expression of serum IL-8 and peripheral mononuclear cells mRNA in two groups of CHB patients were detected using ELISA method and RT-PCR method,and the content of serum ALT and AST enzyme was detected as well using Roche biochemical analyzer. Results ELISA and RTPCR analysis suggested the content of serum IL-8 and the expression of peripheral mononuclear cells IL-8 mRNA were respectively 252. 65 ±62. 04,1. 50 ± 0. 42 in HBe Ag negative CHB patients,and the content of serum IL-8 and the expression of peripheral mononuclear cells IL-8mRNA were respectively 241. 83 ± 49. 88,1. 28 ± 0. 36 in HBe Ag positive CHB patients. There was a good correlation( r = 0. 655) between the content of serum IL-8 and the expression of peripheral mononuclear cells IL-8 mRNA in HBe Ag negative CHB patients,while there was a poor correlation( r = 0. 276) between the content of serum IL-8 and the expression of peripheral mononuclear cells IL-8 m RNA in HBe Ag positive CHB patients. There was a good correlation( r = 0. 489,P < 0. 05) between the content of serum IL-8 and the content of serum ALT enzyme in HBe Ag negative CHB patients. Conclusion There exist certain differences between the expression of IL-8 in HBe Ag negative and HBe Ag positive CHB patients,and IL-8 is associated with the deterioration of liver function in part of CHB patients.
