糖脂平颗粒对糖尿病胰岛素抵抗大鼠胰岛素样生长因子1和内皮素的影响

【摘要】 目的探讨糖脂平颗粒对糖尿病胰岛素抵抗大鼠胰岛素样生长因子1(IGF-1)和内皮素(ET)的影响。方法 Wistar大鼠90只随机选取8只为正常对照组,其余大鼠采用高脂高糖饲料联合注射链脲佐菌素诱导糖尿病胰岛素抵抗大鼠模型。选取造模成功大鼠32只,随机分为模型组、糖脉康颗粒组、糖脂平大剂量组、糖脂平小剂量组,每组8只。正常对照组和模型组给予等体积生理盐水,各给药组每日分别给予糖脉康颗粒(1.35g/kg)、糖脂平颗粒大剂量(2.7g/kg)、糖脂平颗粒小剂量(1.35g/kg),每天灌胃1次,连续4周。检测各组大鼠体重、血糖、血清胰岛素(INS)、IGF-1及ET,并计算胰岛素敏感性指数(ISI)。结果各给药组与模型组比较,体重、血糖、INS及ET均降低,ISI和IGF-1均升高,差异具有统计学意义(P<0.05或P<0.01)。结论糖脂平颗粒可降低糖尿病胰岛素抵抗大鼠体重、血糖、INS和ET,同时升高ISI值和IGF-1,从而改善了糖尿病模型大鼠的胰岛素抵抗。

【Abstract】 Objective To observe the effect and mechanism of Tangzhiping Granules(TZPG) for diabetic rats with insulin resistance(IR).Methods Eight rats among ninety Wistar rats of specific-pathogen free(SPF) grade were selected randomly as the normal control group and 82 diabetic rat models with IR were induced by a high fat,high sugar diet and injection of streptozotocin(STZ).Thirty-two successful modeling rats were randomized into the model group,Tangmaikang Granules(TMKG) group,high-dose TZPG group,low-dose TZPG group,with 8 in each.The same volume of saline was given to the normal control group and model group respectively.The other groups were given intragastric administration of 1.35 g/kg TMKG,2.7 g/kg TZPG and 1.35 g/kg TZPG once daily for 4 weeks respectively.The body weight,blood glucose(BG),serum insulin(INS),insulin-like growth factor-1(IGF-1) and endothelin(ET) were detected and the insulin sensitivity index(ISI) was calculated.Results Comparing with the model group,BG,INS and ET were significantly decreased,but ISI and IGF-1 were significantly increased in the TMKG group,high-dose TZPG group and low-dose TZPG group(P<0.05 or P<0.01).Conclusion TZPG can reduce the body weight,BG,INS and ET,increase ISI and IGF-1 and improve the IR in diabetic rats.
