圆红冬孢酵母发酵玉米秸秆水解液产油脂的研究
Production of microbial oil by Rhodosporidium toruloides with corn straw hydrolysate
【摘要】 以玉米秸秆为原料,圆红冬孢酵母As 2.1389为实验菌株,研究了玉米秸秆稀酸水解的条件及该菌种在水解液中发酵产油脂的特性。结果表明,玉米秸秆的最佳水解条件为:硫酸体积分数2%,固液比1∶10,121℃高压水解20 min;在此条件下,水解液中的还原糖含量为38.77 g/L。圆红冬孢酵母在经Ca(OH)2脱毒处理后的水解液中进行发酵产油脂,最佳发酵条件为:pH 5,接种量15%,发酵时间5 d,得到的生物量为13.5 g/L,油脂产量4.71 g/L,油脂含量为34.89%,还原糖利用率95.73%。气相色谱-质谱分析表明其油脂的脂肪酸组成与植物油脂相似,可作为制备生物柴油的原料油脂。
【Abstract】 With corn straw as raw material and Rhodosporidium toruloides As 2.1389 as test strain,the dilute acid hydrolysis conditions of corn straw and the characteristics of this strain fermentating in the hydrolysate were studied.The results showed that the optimal hydrolysis conditions were as follows:H2SO4 volume fraction 2%,ratio of solid to liquid 1∶ 10,time 20 min,high pressure,temperature 121 ℃.Under these conditions,the reducing sugar content in hydrolysates was 38.77 g/L.The hydrolysates detoxified with Ca(OH)2 were well utilized by the yeast to synthesize biomass.The optimal fermentation conditions were pH 5,inoculum dosage 15%,fermentation time 5 d,and the yield of biomass reached 13.5 g/L,the oil productivity was 4.71 g/L,the oil content was 34.89% and the utilization rate of reducing sugar was 95.73%.The fatty acid composition of the microbial oil was similar to vegetable oil analyzed by gas chromatography-mass spectrometry,so it could be used as raw material for biodiesel.
