乌蕨对S180荷瘤小鼠的抑瘤作用

【摘要】 目的:观察乌蕨水提取物对荷瘤小鼠的抑瘤作用及其对血清超氧化物歧化酶(superoxide dismutase,SOD)、谷胱甘肽过氧化物酶(glutathione peroxidase,GSH-PX)的活性及丙二醛(malondialdehyde,MDA)、一氧化氮(nitric oxide,NO)含量的影响。方法:采用右前肢腋部皮下接种S180腹水瘤方法建立S180荷瘤小鼠模型。于造模后第二天,灌胃给予不同剂量的乌蕨水提取物(2.5,5,10 g/kg)或生理盐水,连续14 d。观察小鼠体重变化,计算瘤重和肿瘤指数、脾脏指数及胸腺指数,测定小鼠血清中SOD、GSHPX的活性及MDA、NO的含量。结果:2.5,5,10 g/kg乌蕨组小鼠肿瘤生长明显受到明显抑制;脾脏指数降低,胸腺指数升高;血清SOD活性升高,以5,10 g/kg乌蕨组效果较好;2.5及10 g/kg乌蕨组小鼠血清MDA含量比模型组降低;2.5 g/kg乌蕨组小鼠血清NO含量高于模型组,与CP组相比,2.5,5 g/kg乌蕨组小鼠血清NO含量升高;2.5 g/kg乌蕨组小鼠血清GSH-PX活性升高。结论:乌蕨水提取物具有抗肿瘤作用,其抑瘤作用可能与提高小鼠的抗氧化能力有关。

【Abstract】 Objective: To observe the anti-tumor effect and its mechanism about antioxidant of Stenoloma chusanum( L.) Ching water extract( SCWE). Methods: S180-bearing mice model were established by subcutaneously inoculating S180 ascitic tumor into the right armpit. Once the model is formed,the mice will be administrated with various doses of SCWE( 2. 5,5,10g / kg) or NS through ig from the second day,and the process will extend 14 days. After the last administration,the mice body weight,tumors weight and the index of tumor,speen and thymus were calculated,and the content of MDA and NO,the activity of SOD and GSH-PX in serum were measured. Results: Compared with the model group,SCWE groups showed inhibitions in tumor growth and spleen index,enhance in thymus index,increased activities of SOD and GSHPX,and decreased contents of MDA and NO. Conclusion: SCWE had an anti-tumor effect,and its mechanism probably result of improving the antioxidant ability of S180-bearing mice.
