灯盏花素对正畸牙移动大鼠牙槽骨重建的影响
Effects of breviscapine on the periodontal tissues remodeling of rats with orthodontic tooth movement
【摘要】 目的:研究灯盏花素对正畸牙移动大鼠牙槽骨重建的影响。方法:56只雄性SD大鼠随机分为对照组和实验组,每组28只,建立大鼠正畸牙移动模型,50g力正畸装置牵引上颌第一磨牙近中移动,隔日实验组每日灌服灯盏花素100㎎/㎏,对照组每日给予同等剂量生理盐水。两组大鼠分别在加力第7、14、21、28天后分批处死,测量各个时间点上颌第一磨牙的移动距离,HE染色法观察牙周组织及压力侧破骨细胞数量变化,免疫组织化学染色法观察牙槽骨中压力侧IL-1β表达变化。结果:正畸加力第7天实验组(灯盏花素100㎎/㎏)大鼠牙移动距离与对照组无显著性差异,破骨细胞数量增加有显著性差异,牙槽骨中IL-1β阳性表达增强,与对照组有显著性差异。加力第14、21、28天实验组大鼠牙移动距离比对照组分别增加0.12mm、0.23mm、0.19mm,破骨细胞数量比对照组分别增加1.27、1.49、1.56,两组比较差异有显著性,IL-1β表达平均灰度比对照组分别增加34.37、33.94、30.91,两组比较差异有显著性。结论:灯盏花素通过上调牙周组织中IL-1β的表达,促进破骨细胞的增殖和分化,有利于正畸牙移动。
【Abstract】 Objective: To observe the effects of breviscapine in the periodontal tissues of rats during orthodontic tooth movement.Methods: 56 healthy male 8-week-old SD rats were selected.They were randomly divided into 2 groups: breviscapine group and control group.Then animal model for orthodontic tooth movement was established.the same daily dose of physiological saline was drenched to the control group and the breviscapine group was given 100 mg / kg per day breviscapine decoction.The rats were executed in batch on the 7th,14th,21st,28th day during the orthodontic treatment.The distances of the tooth movement were measured and slices from the periodontium of the maxillary first molar were observed under optical microscope.Immunohistochemical staining was used to observe the expression of IL-1 beta in alveolar bone in the pressure side of alveolar bone.Measurement data were compared with t test and analysis of variance by SPSS13.0 statistical software.Results: The first seven days orthodontic tooth movement in rats from breviscapine and control group had no significant difference(P > 0.05),the number of osteoclasts was significant difference(P < 0.05),the expression of IL-1 beta in alveolar bone was significant difference(P < 0.05).Compared with blank control group,14th,21st,28th days after burner first experimental tooth movement in rats increased by 0.12mm,0.23mm,0.19mm,respectively;the number of osteoclasts increased by 1.27,1.49,1.56,respectively;the expression of IL1 beta image analysis the average gray level increased by 34.37,33.94,30.91,respectively.The difference was statistically significant(P < 0.05).Conclusion: Breviscapine can promote the proliferation and differentiation of osteoclasts during orthodontic tooth movement in rats by regulating IL-1 beta expression and increasing osteoclast proliferation and differentiation.
