乳鹿颗粒的长期毒性研究
Experimental study on chronic toxicity of Rulu Granule
【摘要】 目的:观察乳鹿颗粒长期灌胃给药对实验大鼠可能产生的毒性反应,为临床应用提供毒理学依据。方法:实验大鼠随机分为对照组、乳鹿颗粒低、中、高剂量组;乳鹿颗粒组分别灌胃给予乳鹿颗粒16.0、48.0和144.0g/kg(分别相当于临床剂量21、64和192倍),每天给药1次,连续给药12周,逐日观察动物行为、外观及大小便,每2周测食物消耗量1次,每周称体重1次,给药12周后和停药恢复2周后每组随机选取雌雄大鼠各5只取血,检测血液学指标、血液生化指标,并计算脏器质量系数,同时对主要脏器进行外观及组织形态学检查。结果:乳鹿颗粒各组大鼠给药及停药恢复期间的一般状况良好,体重增长、摄食情况、脏器系数、血常规、血液生化指标、解剖学和主要脏器的组织形态学检查均正常,与对照组比较无明显差异。结论:乳鹿颗粒无明显的长期毒性,按临床剂量用药是安全的。
【Abstract】 Objective:To observe the chronic toxicity of Rulu Granule,and to provide toxicological basis for clinical application.Methods: Experimental rats were randomly divided into the control groups and Rulu Granule low,moderate and high dosage groups(correspond to Rulu Granule 16.0,48.0 and 144.0g/kg/d,respectively) with 20 rats in each group and half male and half female,equaling to 21,64 and 192 times of clinical estimated dose by p.o.once a day and for 12 weeks.Meanwhile,rats’ behavior and appearance were observed,and night shit and urine everyday amounts were measured.Food intake was measured once two weeks and body weight once every week.The hemotology and biochemistry indexes of 10 rats(randomly,half male and half female) in every group respectively were measured after 12 weeks and recover two weeks after stopping administration,and rats were killed,and organ coefficient were measured and main organs were examined by pathological method.Results: the rats’ behaviors and appearances among the control and three Rulu Granule-treated groups were normal during administration and recovery after stopping administration.The body weight,food intake,main hemotology and biochemistry indexes,organ coefficients of rats in treated groups were basically the same as those of the control.The organ histopathological observation showed no significant difference between experimental rats and control.Conclusion: Rulu Granule has no apparent long-term toxicity,and is safe to use with clinical dosage.
