【摘要】 目的:观察黄连总生物碱(Coptis alkaloids,CA)对糖尿病肾病(Diabetic Nephropathy,DN)大鼠的保护作用。方法:大鼠适应性饲养1周,禁食不禁水12h后,按60mg/kg剂量一次性腹腔注射链脲佐菌素(streptozotocin,STZ)的方法复制大鼠糖尿病模型。72h后尾静脉取血,测血糖,选取血糖值≥16.7mmol/L大鼠100只,随机分为模型组、黄连总生物碱低(50mg/kg)、中(100mg/kg)、高剂量(200mg/kg)组,阳性药格列齐特(20mg/kg)组,另取未造模大鼠10只为正常组。给药组分别灌胃给予黄连总生物碱、格列齐特,正常组和模型组给予等容量溶媒,每天1次,连续8周。末次给药后,禁食12h处置动物,取相应指标检测:检测大鼠空腹血糖(fasting blood glucose,FBG)水平;HE常规染色,光镜下观察大鼠肾脏病理学变化情况;全自动生化仪检测血肌酐(serum creati-nine,Scr)、血尿素氮(boold urea nitrogen,BUN)含量;放射免疫分析法检测胰岛素(Insulin,Ins)、C肽(C-PePtide,C-P)、肿瘤坏死因子(Tumor necrosis factor,TNF-α)的含量。结果:实验结果显示,与模型组相比,黄连总生物碱(50mg/kg、100mg/kg、200mg/kg)组能降低大鼠FBG、TNF-α、BUN、Scr水平,升高Ins;黄连总生物碱(100mg/kg、200mg/kg)组能升高C-P含量;病理组织学检查亦提示,黄连总生物碱能够改善肾脏病理组织学损伤程度。结论:黄连总生物碱对糖尿病肾病大鼠具有一定的保护作用。更多还原

【Abstract】 Objectives:Study the protective effect of Coptis alkaloids on diabetic nephropathy rats. Methods: Model rats was induced by streptozotocin. All the therapeutic groups were treated with Coptis alkaloids ( 50,100,200mg/kg) and the gliclazide (20mg/kg) . Meanwhile,the CMC-Na were used in the normal group and the model group. Above medication administrations were given once a day and lasted for 8 weeks. After 8 weeks administration,the level of serum FBG BUN and Scr were measured by chromatometry. Meanwhile the Ins,C-P and TNF-α were determinated by radioimmunity method. Moreover,kidney samples were taken to examine the degree of damage by HE. Results: As compared with the model group,Coptis alkaloids ( 50mg/kg,100mg/kg,200mg/kg) significantly reduced histopathological change,degraded FBG,Scr,BUN,TNF-α contents,steped up the level of Ins and Coptis alkaloids( 100mg/kg,200mg/kg) significantly steped up the level of C-P. Conclusion: Coptis alkaloids has protective effect on early diabetic nephropathy rats.更多还原
