抑郁模型大鼠海马CA3区突触结构的体视学变化及加味温胆汤的作用

【摘要】 目的:探讨抑郁模型大鼠海马CA3区突触结构的体视学变化及加味温胆汤的作用。方法:以孤养结合慢性不可预见性应激抑郁模型大鼠为研究对象,在实验第7d,14d,21d,28d,检测各组大鼠海马突触结构的体视学变化。结果:在抑郁形成过程中,21d时,与空白组比较,模型组大鼠突触活性区长度、突触后致密物质厚度明显减少,突触间隙宽度增加明显。28d时,差异进一步明显。随着加味温胆汤(12g/kg)治疗周次的增加,抑郁大鼠突触活性区长度、突触后致密物质厚度减少,以及突触间隙宽度上升的趋势得到抑制,与模型组比较,在21d时,有明显统计学差异(P<0.05),28d时,差异增大(P<0.01)。加味温胆汤汤组(12g/kg)与氟西汀组(1.8mg/kg)相比无明显统计学意义。结论:抑郁症可引起海马神经元突触结构损伤,导致出现神经元萎缩,加味温胆汤对此有逆转作用。

【Abstract】 Objective: To probe stereological changes of hippocampal synaptic structure in CA3 region of rat depression model and results after regulating by Jiawei Wendan decoction.Methods: The isolated model control group was living under unpredictable chronic stress.At the 7th,14th,21st,28th day of the experiment,we detected stereological changes of hippocampal synaptic structure of each group.Results: During the progress of depression,there were some tendencies: At the 21st day,comparing with the blank group,the length of active synaptic region in model control group was shorter,the thickness of post-synaptic dense area started to thin out gradually and the synaptic cleft was widen;At the 28th day,there were increasing differences between the model control group and the blank group.With the therapic weeks prolonging by Jiawei Wendan decoction(12g/kg),these tendencies appeared on depression rats can be controlled.Compared to model control group,there existed significant differences at the 21st day(p<0.05);and these significant differences became larger at the 28th day(P<0.01).Conclusion: stereological damages of hippocampal synaptic structure resulting from depression led to neuron atrophy,which could be reversed by Jiawei Wendan decoction.
