大承气汤对大鼠肠缺血再灌注损伤的保护作用

【摘要】 目的:观察大承气汤对大鼠肠缺血再灌注损伤的保护作用,并初步探讨其机制。方法:SD大鼠随机分为假手术组、模型组和大承气汤高、低剂量组(20g/kg、10g/kg)。采用夹闭肠系膜上动脉的方法建立大鼠急性肠缺血再灌注损伤模型,观察造模前1小时、造模后1小时和2小时肠粘膜血流和全血中性粒细胞检测,运用酶联免疫法测定血浆肿瘤坏死因子及白细胞介素6的含量变化。结果:大鼠肠缺血再灌注后,肠粘膜血流明显下降,全血中性粒细胞、血浆肿瘤坏死因子和白细胞介素6显著增高;大承气汤(20g/kg、10g/kg)干预后,可在肠缺血再灌注1小时和2小时后,表现出不同程度的逆转作用。结论:大承气汤可通过促进血流恢复,减轻炎性反应等发挥对大鼠肠缺血再灌注损失的保护作用。

【Abstract】 Objective: To observe the protective effect of Dachengqi decoction against intestinal ischemia-reperfusion injury and its mechanism.Methods: SD rats were randomly divided into sham group,model group,Dachengqi decoction high-dose group(20g/kg)and Dachengqi decoction low-dose group(10g/kg).The establishment of Acute intestinal ischemia-reperfusion injury rats model with occlusion of the superior mesenteric artery.Intestinal mucosal blood flow,whole blood neutrophils,tumor necrosis factor-and interleukin-6 content of the plasma levels using enzyme-linked immunosorbent assay were observed one hour prior to the modeling,modeling in one hour and two hours.Results: After intestinal ischemia reperfusion,intestinal blood flow of rats decreased significantly(P<0.01),whole blood neutrophils,plasma levels of tumor necrosis factor-and interleukin-6 were significantly higher(P<0.01);Dachengqi(20g/kg、10g/kg)decoction intervention showed varying degrees of reversal effect(P<0.05,P <0.01)after intestinal ischemia-reperfusion in 1 hour and 2 hours.Conclusion: Dachengqi decoction played a protective effect against intestinal ischemia-reperfusion damage by promoting the restoration of blood flow,reducing the inflammatory response.
