乳化异氟醚后处理对缺血/再灌注大鼠离体心脏超微结构的影响

Effect of emulsified isoflurane postconditioning on myocardial ultrastructure protection in isolated rat hearts with ischemia/reperfusion injury

【摘要】 目的观察乳化异氟醚(EI)后处理对缺血/再灌注成年大鼠离体心脏超微结构和线粒体评分的影响,并探索EI后处理心肌保护作用的可能机制。方法建立SD大鼠缺血再灌注损伤模型,将建模成功的心脏随机分成8组(n=8):正常组(N)、缺血/再灌注组(I/R)、缺血后处理组(IPO)、乳化异氟醚后处理组(EI)、脂肪乳后处理组(FAT)、MPG+IPO组、MPG+EI组、MPG+FAT组,用透射电子显微镜观察各组心肌组织的超微结构,并进行线粒体评分。结果 EI组、IPO、FAT、N组超微结构损害及评分低于I/R组(P<0.01)。EI组和IPO组心肌超微结构损害及线粒体评分无统计学差异(P=0.545),但均低于FAT组(P<0.01)。应用MPG后,除M+FAT组外,各组心肌超微结构损害及线粒体评分均高于相应的非阻断组(P<0.01)。结论 EI和IPO后处理均能减轻大鼠心肌的缺血再灌注损伤,且两者效果相当;活性氧清除剂能消除EI和IPO的心肌保护效果;ROS是介导EI后处理和缺血后处理心肌保护作用的重要因子。

【Abstract】 Objective To observe the effect of emulsified isoflurane postconditioning on myocardial ultrastructure and mitochondria score in isolated heart of adult rats with ischemia / reperfusion injury,and to explore the possible mechanisms of cardiac protection.Methods The hearts of adult male Sprague Dawley rats were isolated and subjected to ischemia-reperfusion injury.Hearts were divided into eight groups(n = 8,each group),i.e.Normal group(N),ischemia-reperfusion group(I / R),ischemic postconditioning group(IPO),emulsified isoflurane postconditioning group(EI),fat emulsion injection group(FAT),MPG + IPO group,MPG + EI group,MPG + FAT group.The ultrastructure and mitochondria score were examined by transmission electron microscope.Results The ultrastructure damage and mitochondria score of myocardial cells were lower in emulsified isoflurane postconditioning,ischemia postconditioning group,and in fat emulsion injection group as compared with those in ischemia-reperfusion group(P < 0.01).The ultrastructure damage and mitochondria score of myocardial cells in emulsified isoflurane postconditioning group were similar to those in ischemia postconditioning group(P = 0.545),but were lower as compared with those in fat emulsion injection group(P < 0.01).After application of the MPG blocker,the damage and mitochondria score of myocardial ultrastructure in groups except for the fat emulsion injection group were higher than those in the corresponding non-blocking group(P < 0.01).Conclusion Emulsified isoflurane postconditioning and ischemia postconditioning have a similar protective effect inmyocardium from ischemia-reperfusion injury in isolated rat hearts The application of the reactive oxygen species scavenger can abolish the cardioprotective effect on ultrastructure of myocardial cells in emulsified isoflurane postconditioning and ischemia postconditioning.Thus,ROS is an important factor that induces myocardial protection in emulsified isoflurane postconditioning and in ischemic postconditioning.
