电针治疗脑瘫机理的实验研究

【摘要】 目的通过检测缺氧缺血脑瘫模型幼鼠电针治疗后脑组织内乙酰胆碱酯酶(AChE)活性的变化,探讨电针治疗脑瘫的作用机制。方法采用左侧颈总动脉结扎制备缺氧缺血脑瘫模型,按随机分组法分为A组假手术组、B组空白模型、C组针刺治疗组和D组电针治疗组。治疗观察10d为一疗程,疗程结束后断头分离脑组织取皮层、海马、纹状体,离心后取上清,用酶联免疫反应法检测AChE的含量。结果针刺、电针均能提高脑瘫模型幼鼠脑组织内AChE的活性,且电针能显著提高脑瘫模型幼鼠脑组织内AChE的活性。假手术组、电针组与模型组比较幼鼠脑组织内的AChE含量明显升高(P<0.01,P<0.05),在海马部位针刺组与模型组比较幼鼠脑组织内的AChE含量升高(P<0.05),在大脑皮层电针组与针刺组比较幼鼠脑组织内AChE含量升高(P<0.05)。结论电针治疗后缺氧缺血脑型脑瘫幼鼠脑组织内AChE的活性明显升高可能是电针治疗脑瘫的主要作用机制。

【Abstract】 Objective Toinvestigate the changes of brain acetylcholinesterase(AChE) activity in cerebral palsy rat pupstreated by electro-acupuncture and explore the mechanisms of electro-acupuncture treatment in cerebral palsy.Methods Rat hypoxic and ischemic cerebral palsy model was created by left common carotid artery ligation.Rats were randomly divided into sham-operated group,model group,acupuncture group and electro-acupuncture treatment group.Ten days after treatment,rats were sacrificed and the cortex,hippocampus and striatum were separated.The AChElevel was detected by ELISA.Results Compared with the model group,the AChElevel was increased after acupuncture and electro-acupuncture treatment,in which the AChElevel after electro-acupuncture treatment was higher than that after acupuncture treatment.Compared with the model group,the AChElevel in cortex,hippocampus and striatum were increased in sham-operated group and electro-acupuncture treatment group(P < 0.01,P < 0.05).Furthermore,compared with the model group,the AChElevel in the hippocampus after acupuncture treatment was significantly increased(P < 0.05).Compared with the acupuncture group,the AChElevel in the cortex after electro-acupuncture treatment was obviously increased(P < 0.05).Conclusion Electro-acupuncture presents a potential benefit for hypoxic and ischemic cerebral palsy treatment and the mechanisms might be due to the increased brain AChE activity.
