甲硫氨酸合成酶基因A2756G多态性与子痫前期发生的相关研究

【摘要】 目的研究蛋氨酸合成酶(MTR)基因A2756G多态性与子痫前期发生的关系。方法采用聚合酶链反应—限制性片段长度多态性(PCR-RFLP)法对156例子痫前期,126例正常妊娠女性MTR的A2756G进行基因分析。比较上述各组基因型和等位基因频率分布有无差异。结果 MTR基因A2756G突变型等位基因G频率在实验组和对照组中有显著性差异,AG,AG+GG基因型频率分布差异有显著性(P<0.05)。AG基因型是AA基因型发生子痫前期风险的2.73倍,突变基因型是纯合野生基因型生育子痫前期风险的2.65倍。结论 MTR A2756G基因多态性与子痫前期发生相关,基因突变增加了子痫前期的发生风险。

【Abstract】 Objective:To explore the relationship between genetic polymorphisms in methionine synthase gene(MTR),a central enzyme in folate metabolism,and the risk of preeclampsia.Methods: Genomic DNA was isolated from the peripheral lymphocytes of 156 patients with preeclampsia and 126 age matched control subjects.Polymerase chain reaction and restriction fragment length polymorphism were used to examine the polymorphisms of MTR genes A2756G and the relation between these genotypes and risk of preeclampsia was analyzed.Results: It was obvious difference between experiment and control group in MTR gene A2756G mutation allele(G) frequency(P<0.05),AG and AG+GG had obvious differences(P<0.05).The individuals with 2756AG genotype had a 2.73 fold increased risk of developing preeclampsia compared with those who had 2756AA genotype,and A>G had a 2.65 fold increased risk of developing preeclampsia than wild-type Homozygous.Conclusion: MTR gene mutation allele is related to preeclampsia,AG+GG、AG gene mutation type increased the risk of preeclampsia.
