【摘要】 目的检测复方儿茶提取液对人结肠癌HT-29细胞水通道蛋白3(AQP3)表达的影响,初步探讨复方儿茶水提液发挥止泻作用的分子机制。方法实验共分3组,高剂量组、低剂量组和空白对照组。不同剂量处理分为高(40 mg/L),低(10 mg/L)剂量组,分别作用12 h后应用RT-PCR和Western blot法检测高低剂量含药培养液对HT-29细胞AQP3表达的影响。结果 RT-PCR和Western blot结果显示,高低剂量含药培养液均上调HT-29细胞AQP3表达水平。结论复方儿茶水提液可以上调AQP3表达,是发挥止泻作用的重要机制之一。更多还原

【Abstract】 Objective:To investigate the effect of water extraction solution of catechu composita on Expression of Aquaporin-3(AQP3)in HT-29 cells.Methods:Cells were divided into 3 groups:control group,high dose group,low dose group,which were treated with PBS or high/low dose drug medium with water extraction solution of catechu composita for 12 hours respectively.RT-PCR and Western blot were used to detect the effects of high and low dose drug culture medium on the expression of AQP3 in HT-29 cells.Results:RT-PCR and Western blot results showed that AQP3 expression level in HT-29 cells were up-regulated by both high and low dose drug medium with water extraction solution of catechu composita(P<0.05).Conclusion:Water extraction solution of catechu composita can upregulate the expression of AQP3 in HT-29 cells,and play an important role of antidiarrheal.更多还原
