【摘要】 目的:探讨蛋白Z依赖的蛋白酶抑制剂(ZPI)基因外显子-3(exon-3)多态性是否影响妊娠高血压疾病子痫前期患者的凝血功能。方法:选择无血缘关系的健康孕妇40例、妊娠期高血压疾病子痫前期患者48例,用凝固法测定凝血酶原时间(PT)、活化部分凝血活酶时间(APTT)、凝血酶时间(TT)和纤维蛋白原含量(Fg)。用直接DNA测序法对ZPI基因exon-3测序分析。结果:ZPI基因外显子-3在exon-3 137A>G(K46R)、181A>G(S61G)、481A>T(T161S)存在基因多态性;健康孕妇与妊娠高血压子痫前期比较,无论哪种基因突变,其不同基因型之间PT、APTT、TT和Fg结果差异均无统计学意义(P均>0105)。exon-3的181位基因多态性和exon-3的481位基因多态性密切相关,即181位基因型为AA型的个体在481位基因上均为AA型,181位基因型为AG型的个体在481位基因上均为AT型,181位基因型为GG型的个体在481位基因上均为TT型。结论:ZPI基因exon-3 137A>G、181A>G、481A>T多态性并不影响妊娠期高血压疾病子痫前期患者凝血功能,且181A>G基因多态性变化与481A>T基因多态性有相关性。更多还原

【Abstract】 [Objective] To explore the relationship of the exon- 3 polymorphism of protein Z- dependent protease inhibitor( Z PI) gene and the coag ulation function of the preeclampsia of hypertensive disorders in pregnancy. [ Method] The blo od sam- ples were collected from 40 unrelated healthy pregnant women and 48 with the preeclampsia of hypertensive disorders in preg- nancy. The prothrombin time( PT), international normalization ratio( INR), activated partial thromboplastin time( APTT), thrombin time( TT) and fibrinogen( Fg) were measured with solidified method. The exon- 3 DNA sequence of Z PI was detec-t ed by direct DNA sequencing. [ Result] Z PI exon- 3 contains three mutations 137A> G( K46R) 、181A > G( S61G) and 481A > T( T161S). However, when comparing healthy pregnant women with the preeclampsia of hypertensive disorders in pregnan- cy, we found that there were no statistical significance between any of them in PT, APTT, TT, and Fg( P> 0. 05). 181 gene po lymorphism in Exon 3 and 481 gene polymorphism in Exon 3 are closely related, with 181 genotype AA, AG and GG being respectively AA, AT and TT in the 481. [ Conclusion] Z PI gene polymorphisms of exon- 3 137A> G 、181A> G、481A> T do not affect the blood coagulation function of the preeclampsia of hypertensive disorders in pregnancy. The genotype of the 181 gene polymorphism in Exon 3 and 481 gene polymorphism in Exon 3 are closely related.更多还原
