肿瘤临床中中医药治疗的机理研究
Study of Functions of Traditional Medicine in Clinical Tumor Treatment
【摘要】 目的:讨论中药抗肿瘤疗效和Fas/FasL基因对医疗的作用。方法:将肝癌细胞用0、0.01、0.1、1μg/mL表阿霉素一一处理,把上述处理的肝癌细胞和有无参处理的淋巴细胞放在一起培养,观察放入中药或fasl抗体之后对肿瘤的效果;用PCR对化疗药物处理的肝癌细胞Fas/Fasl的具体表达测定,结果:未处理的肝癌细胞OD值最高,而用1μg/mL表阿霉素处理的肝癌细胞OD值高于0,0.01,0.1μg/mL处理的。对于0μg/mL表阿霉素干预的7402肝癌细胞fasl表达对比,7402肝癌细胞表达fas和fasl基因,0.01,0.1μg/mL的降低、1μg/mL的增高;fasl单克隆抗体介入之后,0,0.01,0.1μg/mL表阿霉素处理的肝癌细胞的OD值也高于空白对照。结论:中药抗肿瘤可以增效减毒;Fas/FasL系统在抗肿瘤免疫机制中扮演着重要的角色。
【Abstract】 Objective:To study the effects of traditional Chinese medicine in the tumor treatments and the functions of Fas/FasL genes.Methods:The lung cancer cells were disposed by 0,0.01,0.1,1 μg/mL pharmoribicin and lymphocyte which had or not had astragalus and then the actual antibody functions of tumor treatment with FasL monoclonal or traditional Chinese medicine were observed;by means of PCR,the actual explanation of lung cancer cell Fas/FasL genes was tested.As a result,the OD of lung cancer cell with 1 μg/mL pharmoribicin was higher than that with 0,0.01,0.1 μg/mL pharmoribicin and the highest was in that without pharmoribicin.Compared with those disposed in 0 μg/mL pharmoribicin and Fas and FasL genes in 7402 lung cancer cells,those in 1 μg/mL was increased while those in 0.01,0.1 μg/mL decreased;after the introduction of FasL monoclonal antibody,the OD in lung cancer cells disposed by 0,0.01,0.1 μg/mL pharmoribicin was also higher than the original.Conclusion: Traditional Chinese medicine can reduce the toxicity in tumor treatment and Fas/FasL plays an important part in anti-tumour immune mechanism.
